摘要
目的探讨miR-133b在膀胱癌组织中的表达及其临床意义。方法该研究采用茎环实时荧光逆转录聚合酶链反应(stem-loop real-time,RT-PCR)检测膀胱尿路上皮癌组织和癌旁正常组织中miR-133b的表达丰度,分析miR-133b在不同组织中的表达差异及其与膀胱尿路上皮癌临床病理因素间的关系,探讨miR-133b在膀胱癌发生与进展中的重要作用。结果采用Real-time PCR方法检测41例膀胱尿路上皮癌及配对癌旁正常组织miR-133b的相对表达量,结果显示miR-133b在膀胱尿路上皮癌组织中的表达量较癌旁正常组织明显降低(P<0.001),癌旁正常组织中的表达量是癌组织中的3.02倍。同时发现miR-133b的表达量与肿瘤病理级别存在显著相关(P<0.01),即肿瘤恶性程度越高(高级别组)miR-133b的表达量越低。结论与癌旁正常组织比较,miR-133b在癌组织中呈明显低表达,且与肿瘤病理分级显著相关,提示miR-133b可能在膀胱癌发病过程中发挥重要作用。
【Objective】 To investigate the expression of miR-133b and its correlation with the clinical pathological features in bladder urothelial carcinoma.【Methods】 The expression of miR-133b in bladder cancer tissues and adjacent normal tissues was confirmed by stem-loop real-time PCR,and we analyzed the relationship between the expression and clinicopathological factors of bladder cancer.【Results】 The expression of miR-133b in bladder cancer tissues from 41 patients was significantly down-regulated which were confirmed by stem-loop real-time PCR,of which the expression was 3.02 times higher in the matched control than in tumor(P〈 0.001).According to the statistical results,the lower expression level of miR-133b was strongly associated with high-grade bladder cancer(P〈 0.01).【Conclusions】 The expression of miR-133b in bladder cancer tissues was significantly down-regulated,which was implicated to have close relationship with progression of bladder cancer.
