高含量EPA/DHA甘油三酯型鱼油对小鼠的免疫调节作用 被引量:12

Effects of the high levels of EPA/DHA triglyceride-type fish oil on immune regulation in mice

摘要 目的采用免疫功能低下小鼠模型研究高含量EPA/DHA甘油三酯型鱼油的免疫调节作用。方法Balb/c小鼠,随机分为正常对照组、模型对照组、阳性对照组、乙酯型鱼油组、低含量EPA/DHA甘油三酯型鱼油组和高含量EPA/DHA甘油三酯型鱼油低、高剂量组,每组10只。剂量组小鼠分别灌胃不同种类的鱼油,阳性对照组灌胃左旋咪唑(20mg·kg-1),正常和模型对照组灌胃生理盐水,连续28d。除正常对照组外,其余各组小鼠均于首次给药第21d,连续7d皮下注射氢化可的松(19mg·kg-1),建立免疫功能低下小鼠模型。于末次给药后,分别测定小鼠的各免疫指标。结果高含量EPA/DHA甘油三酯型鱼油可显著改善模型小鼠的血清溶血素水平及抗体生成细胞数,提高迟发性变态反应水平和脾淋巴细胞转化能力,促进腹腔巨噬细胞的吞噬能力和碳廓清能力,提高IFN-γ/IL-4比值,且效果优于低含量EPA/DHA甘油三酯型鱼油和乙酯型鱼油。结论高含量EPA/DHA甘油三酯型可显著改善免疫低下模型小鼠的免疫调节功能,其免疫调节作用可能与其甘油三酯含量及EPA/DHA含量相关。 ObjectiveTo study the effect of high levels of EPA/DHA glyceride-type fish oil on immune regulation in immunocompromized mice. Methods Seventy Balb/c mice were randomly divided into sev en groups: a normal group, a model control group, a positive control group, ethyl ester-type fish oil group, low levels of EPA/DHA triglyceride-type fish oil group and high levels of EPA/DHA glyceride type fish oil group. Different types of fish oil were administrated orally every day to mice in dose groups. Mice in positive control group were given orally with levamisole hydrochloride at a dosage of 20mg·kg^-1. Mice in normal and model control groups were administrated with 0. 9 G NaC1 solution as placebo. All mice were administrated once a day for 28d. On the 21th day, all mice except those in the normal group were injected hypodermally with hydrocortisone at a dosage of 19mg·kg^-1 once a day for 7 days to generate an immunocompromized model. The mice were deprived of food for 12h after the last administration, and immunological indicators were tested. Results The high levels of EPA/DHA glycer-ide-type fish oil had remarkable function in immune regulation. The immune function in high levels of EPA/DHA glyceride-type fish oil group was better than that in the low levels of EPA/DHA triglycer-ide-type group and the ethyl ester-type fish oil group in terms of hemolysin content, quantity of anti body forming cells in vivo, delayed hypersensitivity level, spleen lymphocyte transformation, the phag-ocytic index and rate, the ability of macrophages for carbon particle clearance and IFN-γ/IL-4 ratio. Conclusion The high levels of EPA/DHA glyceride-type fish oil could significantly stimulate the immune function in immunocompromized mice. It suggested that the significant effect on immune regulation cor-related to the higher contents of triglycedide and EPA/DHA in all probability.
