摘要
目的:探讨三种来源的细胞因子诱导的杀伤细胞对人食管癌裸鼠皮下移植肿瘤的抑制作用。方法:建立人食管癌裸鼠皮下移植模型。取32只荷瘤裸鼠平分为4组,食管癌患者自体外周血CIK细胞组、健康人外周血CIK细胞组、脐血CIK细胞组及生理盐水对照组。给予20d对应的注射治疗并观察各组移植瘤生长情况,停药4d后处死裸鼠,称量瘤重,计算抑瘤率,观察肿瘤病理特点。结果:CIK细胞治疗组的肿瘤生长明显受到抑制;瘤重分别为(1.01±0.24)g、(0.95±0.16)g、(0.64±0.22)g、(1.89±0.33)g;抑瘤率分别为46.6%、49.7%、66.2%、0%。各组的肿瘤病理特点均与患者原发肿瘤组织形态一致。治疗组与对照组相比,切片下可见较多淋巴细胞浸润及坏死区。结论:三种来源CIK细胞对人食管癌裸鼠皮下移植肿瘤有明显抑制作用,且脐血来源的CIK细胞抗瘤作用最强。CIK细胞可能成为食管癌治疗的有效免疫效应细胞。
Objective To explore the inhibitory effects of cytokine-induced killer(CIK) cells derived from three sources on human esophageal cancer subcutaneous transplantation tumor in nude mice.Methods Subcutaneous transplantation tumor models of human esophageal cancer in nude mice were constructed.32 nude mice models were divided into 4 groups:the CIK cells from peripheral blood of patients with esophageal cancer group,the CIK cells from peripheral blood of healthy human group,the CIK cells from umbilical cord blood group and the saline control group.The tumor volume was observed after 20 days of therapy.Then all the mice were euthanized,and the tumor weights were measured,the tumor inhibition rates were calculated and the pathologic features were observed.Results CIK cells groups had obvious inhibitory effects on tumor growth;the average weight were(1.01 ± 0.24)g,(0.95 ± 0.16)g,(0.64 ± 0.22)g and(1.89 ± 0.33)g,respectively;and the tumor inhibitory rate were 46.6%,49.7%,66.2% and 0%,respectively.HE stain showed that the morphological characteristics of tumor in 4 groups were consistent with human original tumor.There were more lymphocytes infiltration and necrosis regions in the CIK cells groups than that in control group.Conclusion Three sources of CIK cells had obvious inhibitory effects on human esophageal cancer subcutaneous transplantation tumor in nude mice,especially the CIK cells from umbilical cord blood.CIK cells may be the immune effector cells for the treatment of esophageal cancer.
