摘要
为了明确普通小麦-华山新麦草衍生后代材料中所携带的外源遗传物质,利用SSR标记对普通小麦-华山新麦草衍生后代H18和0841进行了鉴定。SSR标记结果表明,331个SSR标记中有271个在亲本华山新麦草和普通小麦7182间具有多态性;85个在华山新麦草中具有清晰且明亮的特异条带,其中9个标记在H18衍生的8个单株中(2n=51~54)具有华山新麦草特征条带。0841的染色体数目为42条,利用筛选的9个标记对其鉴定,发现定位于小麦3D染色体上的标记Xbarc71在0841中具有华山新麦草的特异条带,同时缺失小麦亲本7182的特异条带,推测0841可能是华山新麦草的3Ns染色体代换了小麦的3D染色体。条锈病抗性鉴定显示,0841中抗至高抗条锈菌条中31号和条中32号混合小种。
In this study,SSR markers were used to identify the genetic materials of Psathyrostachys huashanica(P.huashanica) in wheat-P.huashanica derivatives H18 and 0841.The results showed that among the 331 SSR markers which located on single wheat homoeologous group and used to investigate the polymorphism between common wheat 7182 and P.huashanica,271 markers(81.87%) could display polymorphisms and 85 markers could display the specific bands of P.huashanica clearly and brightly.Nine of the 85 markers showed specific bands among 8 plants of wheat-P.huashanica derivatives H18.Cytological analysis showed that the somatic chromosome number of 0841 was 42.Among the 9 selected markers above,Xbarc71 located in wheat chromosome 3D expressed the specific band of P.huashanica and missed the band of wheat.Based on these results,we infered that wheat chromosome 3D was substituted by P.huashanica chromosome 3Ns in this derivative.The strip rust resistance test indicated that 0841 was moderate-highly resistance to mixed Puccinia striiformis f.sp.tritici races of CYR31 and CYR32.
