中药沙苑子对运动性疲劳大鼠脑组织5-HT及其相关代谢物的影响 被引量:9

Effects of Semen Astragali Complanati on 5-HT and Relevant Substances Levels of Brain in Exercise-induced Fatigue Rats

摘要 目的:通过观察中药沙苑子对运动性疲劳模型大鼠海马、间脑5-HT及其相关代谢物的影响,为沙苑子作为运动保健品的开发利用提供实验依据。方法:采用SD雄性大鼠24只,体重190g~230g,随机分为安静对照组6只、一般训练组6只、强化训练组6只、强化加药组6只,进行7周递增负荷跑台运动建立运动性疲劳大鼠模型,于第8周始运动后即刻取材,运用氨基酸自动分析仪测定大鼠脑组织Trp含量,运用高效液相色谱荧光检测海马、间脑5-HTP、5-HT、5-HIAA含量。结果:1)实验大鼠海马、间脑组织Trp、5-HTP、5-HT含量在耐力跑台运动后增加;随着强度的加大和时间的延长,Trp、5-HTP、5-HT含量增加更为明显,进而加重疲劳程度,沙苑子组大鼠Trp、5-HTP、5-HT含量均显著下降;2)长时间耐力跑台运动后,实验大鼠海马和间脑5-HIAA含量虽高于安静状态和一般训练状态,但未见显著性差异,沙苑子可增强5-HIAA含量的升高。结论:运动可使机体5-HT代谢显著加强,尤其是在长时间大强度运动刺激后,中药沙苑子可通过抑制其合成前体,有效加速5-HT的分解,减少5-HT的积聚,既可推迟运动疲劳的发生,又可及时消除疲劳。 Objective:To provide experimental basis for development and utilization of semen as- tragali complanati as health products, through observing the effects of semen astragali com- planati on 5-HT and relevant substances levels of hippocampus and diencephalon in exercise-in- duced fatigue rats. Methods: 24 male SD rats were randomly divided into 4 groups. They are sedentary group, model group, drill group and treatment group. They were given 7 weeks of endurance treadmill training (speed from 15m/rain gradually increased to 32m/min, 20 to 30 min/d duration with the slope at 0 degree). Trp was tested by amino acid automatic analytical instrument, the concentration of 5-HTP, 5-HT, 5-HIAA were tested by high performance liq- uid chromatography with fluorescence detection. Results : 1 ) The concentration of Trp, 5- HTP and 5-HT in hippocampus and diencephalon of rats increased after treadmill exercise, which led to exercise-induced fatigue, the extent aggravated after long term high intensity treadmill exer- cise. Semen astragali complanati could reduce the concentration of Trp, 5-HTP and 5-HT sig- nificantly compared with drill group. 2) The concentration of 5-HIAA in hippocampus and di- encephalon of rats increased after moderate intensity exercise and high intensity exercise, but both showed no significant differences compared with sedentary group. Semen astragali com- planati could enhance the concentration of 5-HIAA compared with drill group. Conclusion.. Ex- ercise can make the metabolism of Trp strengthened significantly, which makes the increase of the concentration of 5-HT especially after long term, high intensity exercise. Semen astragali complanati could modulate the changes, which could suspend the happen of fatigue and help to eliminate the fatigue, improve rat central nerves performance in long time treadmill running.
