摘要
将带有牛酪蛋白基因caseinB的植物表达载体 pAS -2 ,在大豆自花授粉后 ,用注射法导入大豆受精子房中 .以质粒 pAS -2的 3 5S -Casein -Gus片段为模板 ,随机引物法标记探针 ,对 1 0 3株受体植株后代的DNA进行点杂交和Southern杂交 ,发现其中 1株在点杂交和Southern杂交中均呈阳性 .证明外源基因已整合到大豆基因组中 .
Botanic expressing vector pAS-2 plasmid containing bovine casein B gene was introduced into soybean (Glycine max) after self pollinated by injection method.The total DNA of each progeny of recipient plants was assayed by dot hybridization and Southern hybridization in which 35S Casein Gus fragment from pAS-2 plasmid was used as template to synthesize the probe.One among 103 recipient plants gave positive signals in both of the hybridization.The results demonstrated that the exogenous DNA was integrated into soybean genome.
