摘要
目的:探讨天麻素对血管舒缩异常大鼠模型神经肽、一氧化氮系统的影响。方法:大鼠分5组:溶媒组、多巴胺组、模型组、盐酸氟桂利嗪组、天麻素组,在造模后2h取腹主动脉血测CGRP、ET、SP、β-EP、NO、TNOS、iNOS及cNOS;采用RT-PCR技术检测中脑c-fos mRNA。结果:与溶媒组比较,模型组ET含量降低,β-EP及SP含量升高。与模型组比较,盐酸氟桂利嗪组、天麻素组ET表达均升高;多巴胺组、盐酸氟桂利嗪组、天麻素组β-EP含量均降低;盐酸氟桂利嗪组SP含量降低。各组CGRP、TNOS、iNOS、cNOS比较差异没有统计学意义。模型组NO较溶媒组表达增多,天麻素组较模型组表达降低。模型组中脑c-fos mRNA较溶媒组和多巴胺组表达增多,天麻素组和盐酸氟桂利嗪组c-fos mRNA表达较模型组均降低。结论:天麻素治疗偏头痛作用可能与增高ET表达、降低β-EP与NO的含量有关。
Objective:To explore effect of gastrodin on neuropeptides and nitric oxide system of abnormal vasomotor model rat induced by dopamine and nitroglycerin.Methods: Rats were divided into 5 groups: solvent group,dopamine group,model group,flunarizine hydrochloride group and gastrodin group.It was to take abdominal aortic blood detections of CGRP,ET,SP,β-EP,NO,TNOS,iNOS and cNOS,to detect c-fos mRNA of mesencephalon by RT-PCR 2 hours after modeling.Results: Compared with the solvent group,the content of ET decreased in model group,the contents of β-EP and SP increased.Compared with model group,the expressions of ET in flunarizine hydrochloride group and gastrodin group were increased significantly,the contents of β-EP were decreased in dopamine group,flunarizine hydrochloride group and gastrodin group,the content of SP in flunarizine hydrochloride group was reduced.CGRP,TNOS,iNOS and cNOS in each group had no statistically significant difference.Compared with the solvent group,the content of NO increased in model group,in gastrodin group it decreased compared with that in model group.Mesencephalon c-fos mRNA in model group increased compared with that in the solvent group and dopamine group,while decreased in gastrodin group and flunarizine hydrochloride group compared with that in model group.Conclusion: Gastrodin may play its anti-migraine role through increasing ET expression,decreasing the contents of β-EP and NO.
