清热解毒法基于Bcl-2/Bax等凋亡信号通路调控结肠癌的研究 被引量:11

摘要 目的:观察含清热解毒中药小鼠血清对结肠癌CT-26细胞蛋白Bcl-2、Bax、P53、Caspase-3表达的影响。方法:将BALB/c小鼠随机分为用药A组、用药B组、用药C组和模型D组。A、B、C组分别连续使用解毒三根汤灌胃5、7、10 d后,与D组一起通过眼球取血获得含药血清,并作用于结肠癌CT-26细胞,MTT法测细胞24h、48h、72h的抑制率,与未经鼠血清刺激的正常CT-26细胞分别裂解蛋白后,Western blot法检测Bcl-2、Bax、P53、Caspase-3蛋白的表达量。结果:MTT法结果显示解毒三根汤灌胃10天组,即C组对细胞的抑制率最高,该组Bax、P53、Caspase-3蛋白的表达较正常结肠癌细胞(E组)升高,Bcl-2蛋白表达较正常结肠癌细胞降低,且有统计学意义(P<0.05)。结论:解毒三根汤抑制结肠癌CT-26细胞的生长可能与抑制Bcl-2的表达,促进Bax、P53、Caspase-3的表达量有关。 Objective: To investigate the clinical effectiveness of mice serum with Detoxicated Three-Root Mixture for the expression of colon cancer CT-26's cell protein Bcl-2,Bax,P53,Caspase-3.And to explore the probable mechanisms of tumor cell apoptosis induced by Sangen Decoction.Methods: BALB/c mice were randomly divided into four groups:medicine group A,B and C,Model group D.Mice in group A,B and C were then continuously fed with medicine for 5,7 and 10 days.Blood of mice was taken through eyeball along with group D and then acted on CT-26.Inhibitory rate of CT-26 was observed by MTT method at 24h,48h and 72h.At end of treatment,the expressions of Bcl-2,Bax,P53 and Caspase-3 were observed by western blot method.Results: The results of MTT showed that the group of 10 days made the highest inhibition rates.In this group the protein expressions of Bax,P53 and Caspase-3 were increased compared to those of normal colon cells(group E),and Bcl-2 was lower than that of normal cells(P〈0.05).Conclusion: Detoxicated Three-Root Mixture can inhibit the growth of colon cancer CT-26 cell.The mechanism of it may be related to the apoptosis proteins' expressions of Bcl-2,Bax,P53,Caspase-3,which are adjusted by Detoxicated Three-Root Mixture.
