摘要 目的:筛选大孔树脂吸附法制备小蓟中蒙花苷的最优条件。方法:采用大孔吸附树脂富集小蓟中的蒙花苷,用高效液相色谱法测定样品中蒙花苷的含量,评价6种大孔吸附树脂对蒙花苷的静态吸附/解吸和动态吸附/解吸程度,筛选出对小蓟中蒙花苷富集纯化能力最佳的大孔吸附树脂。结果:经实验,确定6种树脂中的HPD450型大孔吸附树脂对小蓟中蒙花苷的富集纯化能力最佳。其最佳工艺条件为:以3 BV的小蓟供试品溶液(生药浓度为0.5 g.mL-1)为上样量,吸附速率为1 BV.h-1,吸附2 h后,洗脱流速为4 BV.h-1,收集5 BV的水洗脱液,4 BV的30%乙醇洗脱液,3 BV的50%乙醇洗脱液,减压浓缩,即得。结论:用HPD450型大孔吸附树脂从小蓟中纯化得到的蒙花苷含量达75%以上。 Objective:To study the optimal purification technology of linarin from Cirsium setosum by macroporous resin.Methods: HPLC was employed to select the better technique for the purification of linarin from Cirsium setosum by macroporous resin.In the test,the technique was determined on the basis of combining static adsorption with desorption and dynamic adsorption with desorption,as well as the quantity of linarin in elute solution.Results: HPD450 macroporous resin showed good concentration and purification capabilities for linarin from Cirsium setosum.The optimal technology conditions were determined as follows: 3BV decoction of Cirsium setosum(dose of raw drug was 0.5 g·mL-1) was passed through the column at the flow rate of 1 BV/h,and eluted with 5 BV water,4 BV 30% ethanol,3 BV 50% ethanol respectively,elution flow rate was 2 BV/h.30% and 50% ethanol elutes were combined and concentrated to yield extract containing linarin.Conclusion: The purity of linarin was up to 75%.HPD450 macroporous resin was simple and feasible for the purification of linarin from Cirsium setosum.
