德都红花-7味散保肝利胆及调节免疫功能的实验研究

Rharmacodynamic Research on Protecting Liver and Gallbaldder and Immune Regulating Function of Deduhonghua-7 Powder

目的:通过动物实验观察德都红花-7味散对D-GaLN急性肝损伤的保护作用及利胆、调节免疫功能的主要药效学作用。方法:以蒙药制剂"德都红花-7味散"为实验药物,设计高剂量组(0.83 g.kg-1)和低剂量组(0.5 g.kg-1)。实验各组均采用灌胃法连续给药1周。分别采用腹腔注射D-GaLN制作小鼠急性肝细胞坏死模型观察肝功能及肝脏形态学的改变,评价德都红花-7味散对化学性肝损伤的防治作用;采用小鼠腹腔巨噬细胞吞噬鸡红细胞实验,评价德都红花-7味散对巨噬细胞吞噬功能的影响;采用小鼠摘除胆囊直接称重的方法,观察德都红花-7味散对小鼠胆汁分泌的影响;采用MTT法观察徳都红花-7味散对刀豆蛋白A诱导的小鼠脾淋巴细胞增值反应的影响。结果:急性肝损伤实验结果表明,与模型组比较德都红花-7味散组和联苯双酯组均有显著降低ALT,AST作用(P<0.05),与模型组比较德都红花-7味散和联苯双酯组肝组织病理学改变明显减轻。巨噬细胞吞噬功能实验结果表明,与空白组比较德都红花-7味散组和香菇多糖组吞噬率和吞噬指数明显升高(P<0.05),与香菇多糖组比较徳都红花-7味散高剂量组吞噬率明显升高(P<0.05)。利胆实验结果表明,与空白组比较徳都红花-7味散组测定时间内胆汁分泌量升高(P<0.05)。小鼠脾淋巴细胞增值实验结果表明,与空白组比较徳都红花-7味散组OD值升高(P<0.01)。结论:德都红花-7味散对D-半乳糖胺(D-GlaN)所致小鼠急性肝损伤具有保肝降酶作用;能够明显增加小鼠胆汁分泌量;明显增强小鼠腹腔巨噬细胞吞噬功能;对ConA诱导的淋巴细胞增值有促进作用。

Objective：According to animal experiments,it is to observe the protecting function and main pharmacodynamics effects as protecting liver and gallbladder and immune regulating function of Deduhonghua-7 powder on D-GaLN-induced acute liver injury.Methods：The mice of acute liver cell necrosis model was made by intraperitoneal injection of D-GaLN to observe the liver function and morphology changes of liver,in oder to evaluate the chemical protecting function of Deduhonghua-7 powder objectively.The peritoneal macrophages phagocytosis of chicken erythrocytes was used to observe the influences of Deduhonghua-7 powder at phagocytosis function of morphology;the gallbladder removing and weighting method was used to observe the influence of Deduhonghua-7 powder at secretion of bile;MTT method was used to observe the influences of Deduhonghua-7 powder at mouse spleen,lymph cell proliferation induced by concanavalin A.Result：At acute liver injury experiment,comparing with model group,ALT and AST of mice serum in the acute liver injury group and Bifendate group were increased（P〈0.05）.Comparing with model group,Deduhonghua-7 powder group and Bifendate group＇s tissue pathological changes were obviously decreased.At macrophage phagocytosis experiment,comparing with blank group,phagocytosis rate and phagocytosis indexes in Deduhonghua-7 powder group and Lentinan group were significantly increased（P〈0.05）,comparing with Lentinan group,phagocytosis rate of Deduhonghua-7 powder group was significantly increased（P〈0.05）.At choleretic experiment,comparing with blank group,the secreting of bile in determined time in the Dedhuhonghua-7 powder group was increased（P〈0.05）.At mice spleen,lymph cell proliferation experiment,comparing with blank group,the OD value of Deduhonghua-7 powder group was increased（P〈0.01）.Conclusion：Deduhonghua-7 powder has hepatoprotective and enzyme decreasing function on mice with acute liver injury induced by D-GaLN;it can increase mice bile secretion significantly,significantly increase the abdominal macrophage phagocytosis function and improve the proliferation of lymph cell induced by ConA.
