摘要
本文报道了应用表面增强技术将薄层色谱 (TLC)与傅立叶变换拉曼光谱 (FT Raman)联用 ,获得了中草药钩藤中的钩藤碱光谱研究的新方法 .研究表明 ,在薄层原位 2 5μg钩藤总碱 ,即可很好的分离出钩藤碱等多种钩藤生物碱 .应用薄层原位表面增强 (TLC FT SERS)技术 ,获得钩藤碱分子的特征振动谱带 ,进而阐述了样品分子在表面增强基底银胶表面的吸附模式 ,钩藤碱以分子中甲氧基O原子的孤电子对吸附于银晶体微粒表面 ,1397cm- 1的OCH3的变形振动获得最大增强 .说明TLC SERS结合对中草药化学成分进行高灵敏度示踪指纹检测的可靠性和优越性 .
A new method for analysing the ingredients of Rhynchophylline in Uncaria rhynchophylla Jacks using hyphenaed high efficiency thin layer chromatographic (TLC) and the surface\|enhanced raman spectroscopic (SERS) technics is reported.The results show that the characteristic spectral bands of Rhynchophylline can be obtained,the TLC spot with minimum amount of Rhynchophylline sample is about 2.5?μg,the differences between the SERS and solid spectra are found.1?397?cm -1 spectral band is greatly enhanced.Molecules were absorbed in surface of silver sol by pair of electrons in —OCH\-3.An absorption model of Rhynchophylline and silver sol is proposed.The effective combination of TLC and SERS can be used to analyse the chemical ingredients with high sentivity.
