等基线覆盖融合结合相对校正因子对柴胡抗溃疡有效组分的质量控制方法研究 被引量:7

摘要 目的:明确柴胡中柴胡皂苷为抗溃疡有效组分,在此基础上利用等基线覆盖融合结合相对校正因子法,建立柴胡中抗溃疡有效组分-柴胡皂苷质量控制方法研究,为柴胡皂苷抗溃疡临床用药提供依据。方法:通过灌胃无水乙醇的方法建立大鼠胃黏膜损伤模型,评价柴胡皂苷的抗溃疡活性;针对柴胡中抗溃疡活性成分柴胡皂苷采用等基线双波长覆盖融合的方法,对柴胡皂苷进行质量控制研究,在210 nm、254 nm波长下的成分进行融合,外标法测定柴胡皂苷A、柴胡皂苷B2、柴胡皂苷C和柴胡皂苷D的含量;以柴胡皂苷A为内标物,建立柴胡皂苷B2、柴胡皂苷C和柴胡皂苷D的相对校正因子,结合等基线双波长覆盖融合法测定3种成分含量,比较2种方法测定结果的差异。结果:柴胡皂苷可以明显缩小溃疡面积;等基线双波长覆盖融合结合相对校正因子法与外标法测定结果比较无显著性差异。结论:柴胡皂苷对大鼠胃溃疡模型有明显抑制作用;采用等基线覆盖融合结合相对校正因子法可用于柴胡中抗溃疡有效组分-柴胡皂苷的质量控制,为中药质量控制方法的改进提供了新的方法。 Objective: To define the saikosaponin as an ulcer effective component in Bupleuri Radix saponins,and establish quality control method for Bupleuri Radix saponins,so as to provide a basis for the antiulcer clinical use of Bupleuri Radix saponins.Methods: A rat model of gastric mucosal injury was established with ethanol administration by gavage,and the antiulcer activity of saikosaponin was evaluated.Quality control study was performed based on the active anti-ulcer ingredient saikosaponin using baseline dual-wavelength coverage fusion method.The components were fused at wavelength of 210 nm and 254 nm,and the contents of saikosaponin A,saikosaponin B2,saikosaponin C and saikosaponin D were determined using external standard method.Using saikosaponin A as an internal standard,the correction factors of relative saikosaponin B2,saikosaponin C and saikosaponin D were established.Combined with baseline dual wavelength coverage fusion method,the determination results of three ingredients were compared between two methods.Results: Saikosaponin could significantly reduce the ulcer area and its therapeutic effect was almost equal with omeprazole.There was no significant difference between the determination results of baseline dual wavelength coverage fusion method and external standard method.Conclusion: Saikosaponin has a conspicuous restrain effect in rat model of gastric stress ulcer.Quality control of the antiulcer valid component in Bupleurum,saikosaponin,can be achieved using baseline wavelength coverage with a relative correction factor method,which provice a new method for improvement of Chinese medicine quality control.
