p57^kip2 mRNA及蛋白表达水平与宫颈癌发生发展的相关性

摘要 目的检测细胞周期抑制因子p57^kip2 mRNA及蛋白在宫颈癌中的表达水平,探讨其与宫颈癌发生发展的关系。方法用Real-timePCR法检测37例宫颈癌组织和37例癌旁宫颈组织中p57^kip2 mRNA水平;用WesternBolt法检测30例宫颈癌组织和30例癌旁宫颈组织中p57^kip2 蛋白水平;分析p57^kip2 mRNA及蛋白表达与宫颈癌临床病理相关因素的关系。结果宫颈癌组织中p57^kip2 mRNA的表达水平明显低于癌旁宫颈组织(t=9.099,P〈0.01),在宫颈癌组织中p57^kip2 蛋白水平明显低于相应的癌旁宫颈组织(t=6.320,P〈0.01),并且p57^kip2 mRNA及蛋白水平在Ⅰ+Ⅱ期宫颈癌患者显著高于Ⅲ+Ⅳ期宫颈癌患者(tmRNA=6.670,t蛋白=2.314,均P〈0.05)。结论p57^kip2 mRNA及蛋白水平在宫颈癌组织中的表达显著降低,并且随着病理分期的进展其表达进一步下降,提示宫颈癌的发生和发展与p57^kip2 在mRNA及蛋白水平的表达下降有关。 Objective To evaluate the expression levels of p57^kip2 mRNA and p57^kip2 protein in cervical carcinoma, so as to explore their role in the occurrence and development of cervical carcinoma. Methods The levels of p57^kip2 mRNA in 37 cases were evaluated in carcinoma tissues and adjacent non-cancerous tissues with Real-time PCR method. Moreover, the levels of p57^kip2 protein in 30 cases were detected by Western blot in cervical carcinoma tissues and adjacent non-cancerous tissues. The mutual relationships between elinicopathologic factors and p57^kip2 mRNA as well as protein were analyzed. Results Real-time PCR revealed that the level of p57^kip2 mRNA decreased in cervical carcinoma compared to that in adjacent non-cancerous tissues( t = 9. 099, P 〈 0.01 ). The results of Western blot further proved that the level of p57^kip2protein in cervical carcinoma was significantly lower than that in adjacent non-cancerous tissues (t = 6.320,P 〈 0.01 ). Meanwhile, the levels of p57^kip2 mRNA and protein in cases of Ⅰ+Ⅱ stage were significantly higher than those of Ⅲ+Ⅳ stage ( tmRNA = 6. 670, tprotein = 2. 314, both P 〈 0. 05 ). Conclusion In cervical carcinoma, the levels of p57kila mRNA and protein are significantly decreased. With the progress of pathological staging, the levels decrease further, which shows that the occurrence and develooment of cervical carcinoma might be associated with the decreased levels of p57^kip2 mRNA and protein.
