摘要 目的:采用超高效液相色谱-电喷雾离子化串联质谱联用(UPLC-ESI-MS-MS)及傅里叶变换红外光谱(FTIR)技术对一清胶囊化学成分进行分析,并初步探讨其体外抗炎活性。方法:采用ESI离子源,正离子扫描模式下采集数据,并对一级质谱、二级质谱信息进行分析;采用FTIR技术测定10批次一清胶囊光谱图,并进行主成分聚类分析,比较不同批号样品的差异;采用脂多糖(LPS)诱导的RAW 264.7小鼠巨噬细胞模型,观察一清胶囊对巨噬细胞释放NO的影响。结果:根据一级质谱、二级质谱信息及相关文献共推测了12个化合物;FTIR主成分聚类分析表明批号为120703的样品与其他批次样品存在明显差异;体外抗炎实验表明一清胶囊在1~100μmol/L范围内能显著降低细胞上清液中NO的含量,且呈一定的剂量依赖性。结论:UPLC-ESI-MS-MS与FTIR技术为一清胶囊的质量控制提供了快速、简便、有效的手段;一清胶囊具有一定的体外抗炎活性。 Objective :To study the chemical components in Yiqing capsule by UPLC-ESI-MS-MS and its anti-inflammatory activity in vitro. Methods:The mass spectrometer equipped with an eletrospray ion source in positive ion mode was used and data of mass spectrum and mass/mass spectrum were analyzed;FTIR was used to detect the characteristic spectra of ten bcathes of Yiqing capsule. Compared the spectra by principal component analysis and cluster analysis ; RAW264.7 cell model induced by LPS was established to study the inhibitory effect of Yiqing capsule on the release of NO. Results:Totally twelve chemical components were analyzed by the established UPLC-ESI-MS-MS method;The results of the principal component analysis and the cluster analysis indicated that there were significant difierences between the sample(batch number:120703) and other different batches of samples;Yiqing capsule could inhibit the release of NO induced by LPS in a dose-dependent manner at the concentration of 1 -100μmol/L. Conclusion:UPLC-ESI-MS-MS and FTIR which are rapid, convenient and effective, can be used for quality control of Yiping capsule; Yiqing capsule exhibites anti-inflammatory activity in itro.
