采用斑马鱼模型评价甘草酸的抗内毒素作用 被引量:4

Evaluated the antiendotoxin effects of glycyrrhizic acid with zebrafish model

摘要 目的:观察不同浓度的甘草酸对内毒素引起的斑马鱼致命性感染的保护作用。方法:取发育正常的受精后4 d(4dpf)的斑马鱼幼鱼,经卵黄显微注射LPS(2 ng/条)建立内毒素感染模型。以不同浓度的甘草酸处理上述幼鱼,分别于处理后24h、48 h、72 h观察幼鱼的表型和死亡率的变化。结果:与模型组相比,24 h后不同浓度的甘草酸均能减轻斑马鱼的感染,降低死亡率(P<0.01);48 h后,模型组死亡率为100%,0.625 mg/ml甘草酸组死亡率为47.9%,1.25、2.5 mg/ml甘草酸组死亡率均低于23%;72 h后,0.625 mg/ml甘草酸组死亡率增加到59.37%,1.25 mg/ml甘草酸组死亡率为25.54%,2.5 mg/ml甘草酸组死亡率为19.61%。结论:甘草酸对内毒素引起的斑马鱼幼鱼致命性感染有明显的保护作用,且斑马鱼存活率与甘草酸的浓度正相关。 Objective: To investigate the protective effects of glycyrrhizic acid of various concentrations on the fatal infection of zebrafish caused by endotoxin.Methods: At 4 day after fertilization(dpf),the zebrafish larvae with normal development were injected(2 ng/larva) LPS into the yolk for modeling.Then,the larvae were treated with various concentration of glycyrrhizic acid by microinjection and the changes of larval phenotype and mortality were observed at 24 h,48 h,and 72 h after treatment,respectively.Results: After 24 h,compared with model group,various concentration of glycyrrhizic acid could alleviate infection and reduce mortality(P 0.01);After 48 h,model groups' mortality was 100%,0.625 mg/ml glycyrrhizic acid groups' mortality was 47.9%,1.25 and 2.5 mg/ml glycyrrhizic acid group were less than 23%;0.625 mg/ml glycyrrhizic acid group mortality increased to 59.37% after 72 h,1.25 and 2.5 mg/ml glycyrrhizic acid groups' mortality were 25.54% and 19.61%,respectively.Conclusion: Glycyrrhizic acid could protect zebrafish against a lethal challenge with endotoxin in a dose-and time-dependent manner.
