三七总皂苷对大鼠急性心肌梗死的保护作用 被引量:19

Protected effects of Panax notoginseng saponin(PNS)on acute myocardial infarction rats

摘要 目的:通过观察三七总皂苷(Panax Notoginseng Saponin,PNS)对大鼠急性心肌梗塞(AMI)后血流动力学及血清酶的影响,探讨PNS对AMI大鼠保护作用及可能机制。方法:成年雄性SD大鼠40只随机分为4组:假手术组、急性心梗模型(AMI)组、三七总皂苷(PNS,20mg/kg)组、地尔硫卓组(22.5mg/kg)。采用麻醉开胸结扎冠状动脉左前降支造成AMI模型,观察AMI后各组大鼠血流动力学,左心室收缩峰压均值(mlVSP)、左心室舒张压均值(mlVDP)、左心室收缩压最大上升速率(+dp/dtmax)、左心室舒张压最大下降速率(-dp/dtmax)、左心室开始收缩至达到室内压最大上升速率时间(t-dp/dtmax)及心电图、心律失常、血清游离脂肪酸(FFA)和天冬氨酸氨基转移酶(AST)、以及血清肌酸激酶(CK)、乳酸脱氢酶(LDH)、丙二醛(MDA)、超氧化物歧化酶(SOD)、心梗面积的变化。结果:大鼠AMI后,mlVSP、±dp/dtma明显降低,mlVDP明显升高(P<O.O5)。与AMI组相比,PNS(20mg/kg)组显著提高mlVSP(结扎及结扎后30分钟)at the time of ligation and the 120 minutes after ligation、+dp/dtmax(结扎及结扎后120分钟)、-dp/dtmax(结扎及结扎后30分钟)(P<O.O5),显著降低心律失常发生次数(P<0.05)、心肌梗死面积和血清MDA、CK、LDH的含量(P<0.05)及T波的高抬(P<0.05)。结论:PNS能显著改善左室收缩和舒张功能,降低血清酶的含量及心律失常发生率,这可能是其保护心肌梗死的机制之一。 Objective: To research the effects of panax notoginseng saponin(PNS) on hemodynamic and serum enzymes in rats with acute myocardial infarction(AMI) and to discuss the mechanism of PNS treated AMI.Methods: AMI model was induced by left coronary artery ligation.Forty healthy Spraugue-Dawley male rats were randomly divided into sham operated group,AMI group,PNS treated group and Diltiazem treated group.The cardiac function index such as mean Left ventricular systolic(mlVSP),mean Left ventricular diastolic blood pressure(mlVDP),maximal rising and falling rate of ventricular pressure(±dp/dtmax),the time from beginning of Left ventricular systolic to maximal rising rate of ventricular pressure(t-dp/dt) and the area of myocardial infarction,the changes of T waveform,AMI area and serum SOD,MDA,LDH,CK,AST,FFA levels were measured.Results: After AMI,the level of HR,mlVSP and±dp/dtmax were lower than those before AMI,the levels of mlVDP increased significantly(P0.05).Along with the prolongation of ischemia all indexes tended to normal level.Compared to the AMI group,PNS treated group significantly increase mlVSP(at the time of ligation and the 30 minutes after ligation),+dp/dtmax(at the time of ligation and the 120 minutes after ligation),-dp/dtmax(at the time of ligation and the 30 minutes after ligation)(P0.05).Moreover,PNS treated group reduced T waveform level,AMI area,frequency of arrhythmia and serum CK,LDH and MDA levels(P0.05) caused by AMI.Conclusion: PNS reduced T waveform,frequency of arrhythmia,myocardial infarction area and serum CK,LDH and MDA levels and improved the compliance of ventricular wall and significantly improve systolic and diastolic function caused by AMI,which may be one of the mechanisms of PNS against AMI.
