miR—10a在胃癌组织中的表达及意义

Clinical significance of miR-lOa expression in gastric cancer tissues

摘要 目的 探讨miR-10a在胃癌组织中的表达及与临床病理特征的相关性.方法 采用茎环实时RT-PCR方法检测92例胃癌组织及20例正常胃组织中miR-10a的表达,分析miR-10a的表达与胃癌常见临床病理特征的关系.结果 用茎环实时RT-PCR方法检测miR-10a表达的敏感性和特异性良好.miR-10a在胃癌组织中的表达(11.073±1.874)高于正常胃组织(1.083±0.126),差异有统计学意义(P<0.01).miR-10a在有淋巴结转移的胃癌组织中的表达(12.893±1.946)高于无淋巴结转移的胃组织(8.249±1.757,P<0.001);在TNM分期Ⅲ~Ⅳ期胃癌组织中的表达高于Ⅰ~Ⅱ期,差异有统计学意义(P<0.01).miR-10a的表达与性别、年龄、肿瘤分化程度无显著关系(P>0.05).结论 miR-10a在胃癌组织中的表达高于正常组织,与胃癌淋巴结转移和临床分期有一定关联.miR-10a可能在胃癌的发展过程中发挥重要作用,有望成为胃癌治疗的新靶点. Objective To explore the association between the miR-10a expression in gastric cancer tissues and the elinicopathologic features of gastric cancer. Methods Expression of miR- 10a in 92 specimens of gastric cancer tissue and 20 specimens of normal gastric tissue was examined by stem-loop real- time RT-PCR. The association between miR-10a expression level and clinicopathologic features of gastric cancer was analyzed. Results The stem-loop real-time RT-PCR was sensitive and specific in miR-10a detection. The expression of miR- 10a in gastric cancer tissues ( 11.073± 1.874) was significantly higher than that in normal gastric tissues (1.083± 0.126)(P〈0.01) , significantly higher in gastric cancer tissues with lymph node metastases (12.893±1.946) than that in those without lymph node metastases (8.249±1.757) (P〈 0.01), and significantly higher in cases of TNM stages III-IV as compared to those of TNM stages I- [I (P〈 0.01). However it was not significantly associated with gender, age or differentiation of cancer (P〉0.05). Conclusions MiR-10a expression is higher in gastric cancer tissues than that in normal gastric tissues, and is associated with lymph node metastases and TNM stage of gastric cancer. MiR- 10a may play an important role in the development of gastric cancer and may be a promising target for gastric cancer therapy.
