摘要
目的:分析微RNA-466(microRNA-466,miR-466)在宫颈癌、宫颈上皮内瘤变(CIN)及正常宫颈的表达情况及其与临床病理之间的关系。方法:采用茎环RT-qPCR分析23例宫颈癌、47例CIN,以及13例正常宫颈组织中miR-466的表达,并分析其表达与宫颈癌常见的临床病理特征间的关系。结果:茎环RT-PCR方法能特异检测miR-466;荧光定量PCR分析示miR-466在宫颈癌中的表达低于CIN及正常宫颈组织(P<0.05);正常宫颈与CIN之间比较差异无统计学意义(P>0.05);miR-466的表达与年龄、肿块大小、大体类型及组织分化程度差异无统计学意义(P>0.05);miRNA-466与预测的HPV特异性miRNA同源性分析提示,miR-466与HPV16、18的长控制区(LCR)序列高度同源;生物信息学分析提示,miR-466可调控多种靶基因。结论:miR-466在宫颈癌、CIN及正常宫颈中表达存在明显差异,其在正常宫颈及CIN中相对表达量明显高于宫颈癌;miR-466的异常表达,可能与HPV的LCR区高度同源性相关;miR-466可能调控多种靶分子,参与宫颈癌发生发展过程。
Objective: To analyze the expression of miR-466 in cervical cancer, cervical intraepithenal neoplasia and normal cervical tissue specimens, and study its association with clinicopathologic features of cervical cancer. Methods: The expression levels of miR-466 in 23 cervical cancer, 47 cervical intraepithenal neoplasia (CIN) and 13 non-tumor tissue specimens were examined by stem-loop real-time RT-PCR, and the relationship between the expression levels of miR-466 and related clinicopathologic feature of cervical cancer were further studied. Results: The stem-loop RT-PCR was specific enough to detect miR-466; by the realtime-PCR, we conclude that expression of miR-466 in cervical cancer was significantly lower than that in the CIN and normal cervical tissue specimens (P〈0.05). However, there was no obvious difference between CIN and normal cervical tissues (P〉0.05). Moreover, there was no significant association was found with other clinicopathologic features age, tumor size, general type, tissue differentiation (P〉0.05); The homology analysis between miR-466 and the predicted miRNA with HPV specificity showed that miR-466 was highly identity to the long control region (LCR) sequence of HPV16 and HPV18; The bioinformatics showed that miR-466 could regulate and control a variety of target genes. Conclusion: Expression levels of miR-466 in the cervical cancer, CIN and normal cervical tissues are significantly different. In addition, the expression levels of miR-466 are obviously higher in CIN and normal cervical tissue specimens than that in the cervical cancer; The abnormal expression of miR-466 may be related to the high homology with LCR of HPV.miR-466 participate in the occurrence or development of cervical cancer by regulate multiple of target gene.
