白藜芦醇调节肝星状细胞活性的实验研究 被引量:1

摘要 目的探讨白藜芦醇(resveratrol,Res)对活化态肝星状细胞(HSCs)增殖、活化的影响及其机制。方法从肝脏分离纯化培养HSCs,DCFH-DA法检测不同浓度Res对HSCs中活性氧的影响,CCK-8比色法检测Res对HSCs增殖的影响,Western blotting检测HSCsα-肌动蛋白(α-SMA)的表达,PCR检测Res对HSCs活性相关基因表达的影响。结果 Res抑制HSCs中活性氧的产生,明显抑制HSCs的活化与增殖(P<0.05),改变了HSCs活性相关基因(大鼠生肌调节因子MyoD、胶原蛋白Ⅲ及胶原蛋白I)的表达(P<0.05)。结论 Res能够抑制HSCs的活化增殖,Res对HSCs的作用可能与抗氧化及抑制MyoD的表达有关。 Objective To investigate the influence of resveratrol on the regulation of hepatic stellate cells(HSCs) in vitro. Methods HSCs were isolated from liver of SD rats. The 5th generation HSCs were cultured on the 96-well plastic plate. The reactive oxygen species in HSCs with resveratrol of different concentration were tested by DCFH-DA kits. The proliferation of HSCs was tested by CCK-8 test kits. Smoothmuscle α-actin (α-SMA) expression of HSCs was evaluated with Western blotting. The activity-related genes were measured with PCR. Results l HSCs were isolated from liver and cultured Successfully. Resveratrol inhibited the generation of the reactive oxygen species. Proliferation and act!vation of HSCs were inhibited by resveratrol (P 〈 0.05). Resveratrol inhibited the expression Of activity-related genes (MyoD, collagen Ⅲ and collagen Ⅰ) in HSCs (P 〈 0.05). Conclusion Resveratrol can inhibit the proliferation and activation of HSCs. Resveratrol maybe has the antioxidative function and could inhibit the expression of MyoD gene,
