摘要
采用基因工程技术 ,将催化ATP合成的酶—ADK的基因克隆进表达载体YEpN181P2获得重组质粒YEpN ADK ,转化酿酒酵母GRF18后 ,ADK基因获得表达 .用重组菌株GRF18(YEpN ADK)发酵合成ATP其产率比对照菌株GRF18提高 75% .
ATP is an important energy carrier in organisms and widely used in clinics But the backward production method leads to the low productivity and poor competitiveress in the market.This report describes the cloning of adenylate kinase,the enzyme catalizing the AMP phosphorization into ATP,gene into expression vector YEpN181P2 giving rise to recombinant plasmid YEpN ADK,transformation of YEpN ADK into S cerevisiae GRF18 and the expressing of ADK gene in GRF18(YEpN ADK).The results of fermentation show that the recombinant strain GRF18(YEpN ADK) produced 75% more ATP than the control strain GRF18.
