摘要
目的探讨α3nAChRs在阿尔茨海默病(AD)中的抗炎作用。方法应用Western印迹,ELISA及实时定量PCR检测各组细胞核因子(NF-κBp65)和炎性趋化因子单核细胞趋化蛋白1(MCP-1),巨噬细胞炎性蛋白-1α(MIP-1α),肿瘤坏死因子-α(TNF-α)的蛋白及mRNA表达情况。结果与对照组比较,淀粉样蛋白(Aβ)处理α3nAChR siRNA SH-SY5Y细胞NF-κBp65、TNF-α蛋白及mRNA表达增加(P<0.05);MCP-1、MIP-1α表达变化不明显。结论 Aβ引起SH-SY5Yα3nAChR siRNA细胞NF-κBp65、TNF-α蛋白及mRNA表达升高,α3nAChR可能具有一定抗炎作用。
Objective To investigate the expressions and changes of nuclear factor(NF)-κBp65,monocyte chemoattractant protein(MCP)-1,macrophage inflammatory protein(MIP)-1α,tumor necrosis factor(TNF)-α in the SH-SY5Y cells with α3 nAChR subunit mRNA silence when the cells were exposed to Aβ and discuss the anti-inflammation of α3 neuronal nicotinic acetylcholine receptors in Alzheimer′s disease(AD).Methods Western blotting,ELISA and real-time PCR were used to determine proteins and mRNA of NF-κBp65,MCP-1,MIP-1α,TNF-α expressions in cells of AD model.Results Compared with controls,the protein and mRNA expressions of NF-κBp65 and TNF-α were obviously raised in the SH-SY5Y cells with α3 nAChR subunit mRNA silence when the cells were exposed to Aβ(P0.05).However,no changes in the protein and mRNA levels of MCP-1 and MIP-1α were found in the cells with α3 nAChR subunit mRNA silence.Conclusions α3nAChRs may antagonize inflammation reaction resulting from Aβ
