杞菊提取液对老年大鼠视皮层γ-氨基丁酸含量的影响

摘要 目的研究杞菊提取液对大鼠视皮层γ-氨基丁酸(GABA)含量的影响。方法采用邻苯二甲醛(OPA)柱前衍生反向高效液相色谱法检测大鼠视皮层中GABA含量,采用比色法测定谷氨酸脱羧酶(GAD)与GABA转氨酶(GABA-T)活性。结果老年大鼠较青年大鼠视皮层GABA含量显著下降,GAD活性明显降低,GABA-T活性显著上升。药物干预后,与老年空白组相比,老年中、高剂量组GABA含量明显升高(P<0.01),老年低、中、高剂量组GABA-T活性显著降低(P<0.01),且高剂量组效果最优。低、中剂量组GAD活力显著上升(P<0.01),高剂量组稍高于空白组,但无显著差异。结论杞菊提取液可提高大鼠视皮层GABA含量和GAD活性,降低GABA-T活性,对老年性视功能衰退有一定的改善或延缓作用。
