木本番茄组织培养快速繁殖技术研究

摘要 以无菌播种取得的无菌苗顶芽为材料,研究基本培养基、激素浓度配比对诱导分化芽、芽的增殖、伸长及生根的影响,水分、温度、基质对试管苗移栽成活率的影响。结果表明:培养基为MS+6-BA 3.0 mg/L+NAA 0.01 mg/L+蔗糖3%有利于诱导芽的形成、培养基为MS+6-BA 2.0 mg/L+NAA 0.01 mg/L+蔗糖3%有利于芽的增殖、培养基为MS+6-BA 0.5 mg/L+蔗糖3%有利于壮芽伸长;1/2MS+NAA 0.2 mg/L+蔗糖1.5%有利于诱导芽生根;在试管苗移栽阶段,基质选用泥炭土、椰糠和珍珠岩混合,于晚春和秋季移栽,可提高试管苗移栽的成活率,使移栽成活率达到90%左右。 Taking buds of plantlets as material, effects of basic mediums, hormone density matching on inducing bud, bud proliferation, elongation and rooting were studied. Influence of moisture, temperature, substrate on the survival rate of plantlets transplanted were tested, too. The results showed that the medium of MS+6-BA 3.0 mg/L+NAA 0.01 mg/L+sucrose 3% was conducive to induce bud formation, MS+ 6-BA 2.0 mg/L+NAA 0.01 mg/L+sucrose 3% was conducive to bud proliferation, MS+6-BA 0.5 mg/L+ sucrose 3% was eondueive to elongation, I/2MS+NAA 0.2 mg/L+ sucrose 1.5% was conducive to rooting. It could improve the survival rate more than 80% of plantlets transplanted by using the matrix consisting of peat, coco peat and perlite in late spring and autumn.
