格尔德霉素体外抑制人肝癌细胞株HepG2增殖并促进凋亡

摘要 目的研究格尔德霉素对人肝癌HepG2细胞增殖及凋亡的影响。方法不同浓度格尔德霉素与人肝癌HepG2细胞共培养,采用MTT法检测增殖抑制率;采用流式细胞术Annexin V法检测细胞凋亡率。结果在0.2、0.4和0.8μmol/L浓度下,格尔德霉素均显著抑制HepG2细胞的增殖,在干预细胞48h后,其抑制率分别为20.36%、29.45%和42.52%,抑制率随药物浓度增加而升高;同时,细胞凋亡率分别为4.82%、24.47%和53.64%,也呈剂量依赖性,与相应对照组比,差异有统计学意义(P<0.05)。结论格尔德霉素可抑制肝癌HepG2细胞增殖并诱导其调亡。 Objective To investigate the effects of geldanamycin on the proliferation and apoptosis in hu-man hepatoma cell lines HepG2. Methods HepG2 cells were incubated with different concentrations of gel-danamycin for 24 to 72 hours. Cell proliferation was determined by MTT assay. Cell apoptosis was detected by flow cytometry using annexin V and propidium iodide. Results Geldanamycin significantly inhibited the prolifera-tion of HepG2 cells at a dose-dependent manner. When compared to the control,the cell proliferation was inhibit-ed by 20.36%,29.45% and 42.52%,respectively after 48h incubation at the concentration of 0.2,0.4 and 0.8μmol/L of geldanamycin,however,the apoptosis of HepG2 cells was increased by 4.82%,24.47% and 53.64%(P〈0.05). Conclusions Geldanamycin inhibits proliferation and induces apoptosis in HepG2 cells,suggesting a therapeutic effect of geldanamycin in hepatoma treatment.
