低聚麦冬多糖的酶法制备及分离纯化

摘要 为得到较纯的低聚麦冬多糖,优化了低聚麦冬多糖的酶法制备条件,比较了不同方法对低聚麦冬多糖粗提物除蛋白的效果,并使用凝胶柱层析法对其进行了分离纯化。确定最佳酶反应条件为:料液比1∶4,温度45℃,pH 5.0,时间12h。醇沉条件为:乙醇体积分数80%,沉淀8h。比较了4种除蛋白的方法,Sevage-TCA联用法蛋白质去除率最高,为81.18%;低聚麦冬多糖损失率最低,为8.39%。除蛋白后的低聚麦冬多糖经Sephadex G-100凝胶柱分离纯化得到3种组分,分子质量均低于水提醇沉法提取的麦冬多糖。 studied. : The enzymatic preparati The optimum enzymatic on and purification of oli condition was 1 : 4 of liq precipitation condition was alcohol concentration 80%0, remove protein, Sevage-TCA has the best efficiency wit polysaccharide loss rate of 8.39%.
