摘要
从白头翁中提取了皂苷作为酶反应底物,使用菌种Absidiasp.P00r产酶催化底物生成产物,使用硅胶柱层析分离纯化底物和产物。底物经体积比8.5∶1.5和8.3∶1.7的氯仿-甲醇洗脱液洗脱得到4种底物皂苷,得率分别为10.6%、13.8%、8.2%、15.3%,HPLC检测其纯度分别为83.94%、81.99%、80.18%、83.40%;产物经体积比8.5∶1.5的氯仿-甲醇洗脱液洗脱得到3种皂苷,得率分别为15.0%、7.1%、10.3%,HPLC检测其纯度分别为93.45%、91.28%、100.00%。
Saponin was extracted from Pulsatilla chinensis and transformed to the enzymatic products by the Al)sidia sp. POOr, then the substrates and enzymatic products were purified by silica gel column. Four kinds of substrate saponin monomer were obtained by the proportion in 8.5 :1.5 and 8.3 : 1.7 chloroform-methanol as mobile phase, of which yields were 10.6%, 13.8%, 8, 2%, 15.3% HPLC detection showed that the purity of all components from substrates were 83.94%, 81.99%,80.18%, 83.40%, respectively. Three kinds of enzymatic product monomer were obtained by the proportion in 8.5 : 1.5 chloroform methanol as mobile phase, of which yields were 15.0%, 7.1%,10.3%, and the purity of products were 93.45%, 91.28%, 100.00%
