高效免疫调节剂促进肉瘤小鼠肿瘤细胞凋亡实验研究

Experimental Research on Inducement of High-Efficiency Immune Modulatorsto the Apoptosis of Tumor Cells in S180 Sarcoma Mice

目的观察高效免疫调节剂对S180肉瘤小鼠肿瘤抑制作用及对凋亡相关蛋白Bcl-2和Bax表达的影响。方法建立S180肉瘤小鼠模型,将30只模型小鼠随机分为模型组、环磷酰胺组(CYT组)、高效免疫调节剂组(HIS组),小鼠治疗10 d后,取肿瘤组织称重,计算瘤重及抑瘤率;采用Western blot法检测肿瘤组织Bcl-2及Bax蛋白表达情况,并计算Bcl-2/Bax比值。结果 HIS对S180肿瘤生长有明显抑制作用;Western blot结果表明HIS能降低Bcl-2蛋白表达,上调Bax蛋白表达,且Bcl-2/Bax明显下降。结论 HIS可通过抑制Bcl-2蛋白表达、促进Bax蛋白表达来诱导S180肿瘤细胞凋亡,进而发挥抑瘤作用。

Objective The aim of this study is to determine if the high concentration of immunomodulatory drugs in- duces the apoptosis of tumor cells in S180 sarcoma mice via apoptosis-associated Protein. Methods $180 sarcoma mice were set up and then randomly divided into three groups ： Model, CYT and HIS. After 10 days of different treatments, the mice are killed and the weight and inhibition rates of tumor in mice were measured. The expression of Bcl-2 and Bax in tu- mor cell were investigated by Western blot. Results HIS can inhibit the tumor cell proliferation, at the same times it down- regulates the expression of Bcl-2 and up-regulates the Bax in S180 sarcoma. Conclusion HIS induces the apoptosis of tu- mor cells in S180 sarcoma mice by the means of suppressing the expression of Bcl-2 and up-regulating the Bax.
