摘要
目的研究小鼠皮肤移植急性排斥期Th17和Treg细胞亚群的变化情况。方法建立小鼠皮肤移植模型,术后1,3,5,7 d观察移植皮肤组织形态学变化,流式细胞术(FCM)分析脾脏单个核细胞Th17和Treg细胞亚群数量。结果同基因组表皮及皮肤附件形态完整,炎症细胞浸润较轻;Th17和Treg细胞亚群数量变化不明显。异基因组可见移植皮片变性坏死,大量炎症性细胞浸润;Th17细胞在移植后逐渐增多,第7天显著高于第1,3,5天(P<0.01),且各时间点的Th17细胞比例均高于同期同基因组小鼠(P<0.01);Treg细胞第7天明显低于第1天和第3天(P<0.05),也显著低于同基因组小鼠各时间点(P<0.05);Th17和Treg变化呈现明显负相关(r=-0.803,P<0.05)。结论小鼠皮肤移植急性排斥反应与Th17和Treg细胞亚群的变化有关,Th17和Treg作为预测排斥反应的观察指标有重要临床价值。
Objective To study the change of Th17 and Treg cells subsets during the initial post-transplant period in mice skin grafting model. Methods We established a mouse model of skin transplantation, on postoperative 1,3,5,7 days, transplant skin tissue morphology change were observed, Th17 and Treg ceils subsets quantity of spleen individual nucleus cell were ana- lyzed by flow cytometry(FCM). Results In isogenic transplantation group, skin and cutaneous were complete, inflammatory cell infiltration were lighter;Th17 and Treg cells subsets quantity changes were not obvious. In allogeneic transplantation group, graft degeneration necrosis were visible, a large number of inflammatory cells infiltrated, Th17 ceils gradually increased after transplan- tation, and were significantly higher on day 7 than day 1,3,5 (P 〈 0.01 ), and were higher than those in mice with isogenome at the same time(P 〈 0.01 ) ;Treg ceils on day 7 was significantly lower than those on day 1 and 3 (P 〈 0.05 ), and significantly lower than those in mice with isogenome each time point(P 〈 0.05) ;Th17 and Treg change presented an obvious negative corre- lation ( r = - 0. 803, P 〈 0.05 ). Conclusion The acute rejection in Mice skin grafting model is related to the changes of Th17 and Treg cells subsets;Th17 and Treg have important clinical value to predict rejection as a observation index.
