Recovery of PHA from Recombinant E.Coli by KOH Solution

用碱消化法从重组大肠杆菌中提取了PHA. 研究了碱液浓度、温度、消化时间及消化过程液固比等参数对产品PHA质量的影响. 提出了二段消化工艺，提高了PHA的产品纯度，所得PHA产品纯度为98.3%，同时较大幅度地降低了PHA的提取成本.

Extraction of PHA from the recombinant E.coli. by the alkali digestion method was experimentally investigated. When 100 g/L of recombinant E.coli cells containing 49.5% of PHA was treated with 0.1 mol/L KOH at 40℃ for 20 min, PHA was recovered with purity of 94.5%. A two stage process of digestion was proposed for the extraction of PHA. By this method, the PHA purity reached 98.3% and the intrinsic viscosity was 1.02 ml/mg.
