维药神香草对哮喘小鼠气道高反应性的抑制作用 被引量:7

摘要 目的探讨维吾尔药神香草对哮喘小鼠气道高反应性的作用及其可能机制。方法选取雌性健康Balb/c小鼠32只,随机分为4组。采用致敏和雾化的方法制备哮喘模型。末次激发后测定气道反应性,并取肺组织测定MMP-9、TIMP-1 mRNA的表达,采用免疫组化法检测肺组织中MMP-9和TIMP-1的表达。结果哮喘组小鼠在吸入不同浓度乙酰甲胆碱时Penh均增加(P<0.05),且RT-PCR检测和免疫组化结果均提示MMP-9表达增加(P<0.05);但地塞米松组和神香草组中Penh及肺组织MMP-9的表达均低于哮喘组(P<0.05)。TIMP-1 mRNA在哮喘组、地塞米松组和神香草中表达增加,但与对照组比较差异无统计学意义(P>0.05)。结论 MMP-9参与了哮喘气道高反应性的产生,维药神香草可通过调节MMP-9/TIMP-1的平衡以达到降低气道反应性的目的。 To evaluate the effects of Hyssopus officinalis L. on airway hyperresponsiveness and its mechanisms, 32 SPF Balb/c mice were randomly divided into normal group, asthma group, dexamethasone group and Hyssopus officinalis L. group. The asthmatic model was established by egg protein sensibilization and long-term inhalation provocation. The changes of the airway resistance were detected after the last time of provocation. At the end of experiment, the expression levels of MMP-9 mRNA and T1MP-1 mRNA in lungs were detected by RT-PCR and the contents of MMP-9 and TIMP-1 in lung tissue were determined by immunohistochemistry. Results showed that Penh and MMP-9 content in lung tissue were increased in model group than those in normal group (P 〈 0.05), while Penh and MMP-9 expression in lung tissue were decreased in each treatment group than those in model group (P〈 0.05). Furthermore, the level of TIMP-1 mRNA was increased in model group and the treatment group, but there were no significant difference as compared with model group (P 〉 0.05). In conclusion, Uigur Herb ttyssopus officinalis L. may decrease the airway resistance by regulating the balance between MMP-9 and TIMP-1.
