摘要
目的研究黄芪等中药多糖干预机体免疫功能的部分机制。方法常规提取黄芪、防风、当归、柴胡、枸杞、地黄、板蓝根、蒲公英、香菇等9味中药多糖组份,分别按大、中、小剂量罐胃给药,制备小鼠含药血浆;选择浓度为20%的含药血浆干预体外培养的小鼠腹腔巨噬细胞(Mφ),并用ELISA法检测M准释放IL-1β、IL-6、IL-8、TNF-α等细胞因子(CK)的水平。结果除柴胡多糖外,其余8种中药多糖含药血浆对IL-1β、IL-6、IL-8、TNF-α中1个或多个细胞因子水平具有明显升高或降低作用(P<0.05或P<0.01),且具有浓度差异性(P<0.05或P<0.01)。结论黄芪等8味中药调节机体免疫功能的机制,可能与这些中药的多糖组份在体内干预巨噬细胞释放IL-1β、IL-6、IL-8、TNF-α等细胞因子有关。
This study designed to evaluate the mechanism of nine Chinese herbs including Radix Astragali, ete in interfering immune function of mice. The polysaccharide of Radix A stragali, Saposhnikovia divaricata, Lycium chinense, Angelica sinensis, Lentinus edod, Rehmannia glutinosa, Herba Taraxaci, Radix Isatidis and Radix Bupleuri were extracted, and then gavaged to mice with different doses for preparing mouse plasma containing drugs. Plasma containing 20%drugs was chosen to treat mouse celiac macrophages in vitro. Then, ELISA was used to detect the levels of cytokines such as TNF-a, IL-l~, IL-6, IL-8 released by the macrophage. The plasma containing polysaccharide of Radix Astragali, Saposhnikovia divaricata, Lycium chinense, Angelica sinensis, Lentinus edod, Rehmannia glutinosa, Herba Taraxaci and Radix lsatidis under certain concentration could obviously enhance or suppress mouse celiac macrophages to release one or more of cytokines (P〈 0.05 or P〈 0.01) in a concentration -dependent manner (P 〈 0.05 or P 〈 0.01). These results suggested that mechanism of the polysaccharide intervening in the immune function may be partly through stimulating cytokines release.
