妊娠晚期流感病毒感染对后代小鼠肺发育的影响

Effect of maternal influenza virus infection during late gestation on lung development in postnatal BALB/c mice

摘要 目的:研究妊娠晚期流感病毒感染对后代小鼠肺组织形态和肺表面活性蛋白(surfactant protein,SP)-B、-C表达的影响。方法:用非致死量流感病毒A/四川/SWL1/2009 H1N1稀释液滴鼻感染孕15 d BALB/c孕鼠,待其自然生产,取后代小鼠。称取各时点小鼠体重及肺组织重量;观察小鼠肺组织病理学改变;Western blotting法检测肺组织SP-B、-C表达的变化。结果:与对照组相比,感染组小鼠体重明显减轻,肺组织重量轻度增加,肺泡发育程度较低,肺泡隔厚度明显增加,辐射状肺泡计数(radial alveolar counting,RAC)明显减少,肺泡Ⅱ型上皮细胞数量无明显改变;SP-B的表达量在出生当天(P0)降低,在出生后4、7和14 d均明显升高,SP-C的表达在P0升高,在其它各检测时点显著降低。结论:妊娠晚期流感病毒感染可影响后代肺发育,改变后代肺组织SP-B、-C的表达模式,可能对后代肺功能产生深远影响。 AIM: To investigate the effect of maternal influenza virus infection during late gestation on postnatal lung development and the expression of lung surfactant proteins(SP)-B,-C in BALB / c mice.METHODS: Pregnant BALB / c mice on the 15th day of pregnancy were intranasally inoculated with a sublethal dose of influenza A / Sichuan / SWL1 /2009(H1N1,SC /1).Pathomorphology of the lungs in the offspring was observed with hematoxylin-eosine staining.The expression of SP-B and SP-C was detected by Western blotting.RESULTS: Compared with control group,the virusexposed offspring had lower body weight and higher lung weight.The histopathology of the lung tissues showed focal alveolar septal widening with pneumocyte hyperplasia along with alveolar cell degeneration.In addition,the radial alveolar counting(RAC) was significantly lower.The results of Western blotting demonstrated the expression of SP-B in the lung tissues on the birthday(postnatal day 0,P0) decreased,but increased on postnatal days 4,7 and 14.The expression of SP-C was higher on P0 and lower on other time points.CONCLUSION: Maternal influenza virus infection results in diminished postnatal lung growth and alters the expression of SP-B and SP-C,which may have a profound influence on the pulmonary functions.
