松龄血脉康胶囊对自发性高血压大鼠PI3K/Akt信号通路的调节机制探讨 被引量:8

Mechanism of regulatory effect of Songling Xuemaikang capsules on PI3K/Akt signaling pathway in spontaneously hypertensive rats

摘要 目的:探讨松龄血脉康胶囊对自发性高血压大鼠PI3K/Akt信号通路的调节机制。方法:24只10周龄自发性高血压大鼠(SHR)随机分为空白组、中药组和西药组,每组8只。中药组灌胃松龄血脉康20mg/(kg.d);西药组灌胃雷米普利1mg/(kg.d);空白组给予同体积的生理盐水。每周测血压1次,4周后检测肝脏中PI3K、Akt mRNA的表达和Akt的蛋白含量。结果:给药4周后,中药组和西药组肝脏PI3K mRNA表达均明显上调,与空白组比较差异有统计学意义(P<0.01);中药组和西药组肝脏Akt mRNA表达也都上调,与空白组比较差异有统计学意义(P<0.05);肝脏Akt的蛋白含量,中药组和西药组均高于空白组,差异均有统计学意义(P<0.05)。结论:松龄血脉康有可能通过激活PI3K/Akt信号通路,维持SHR大鼠心脏正常的收缩和舒张功能,抑制高血压对心肌的不利影响。 To investigate the mechanism of the regulatory effect of Songling Xuemaikang capsules on the PI3K /Akt signaling pathway in spontaneously hypertensive rats(SHRs).Methods:Twenty-four 10-week-old SHRs were randomly divided into blank group(n = 8),traditional Chinese medicine(TCM) group(n = 8),and Western medicine group(n = 8).The TCM group received Songling Xuemaikang capsules(20 mg/kg.d) by gavage;the Western medicine group received ramipril(1 mg/kg.d) by gavage;the blank group was given an equal volume of saline.Blood pressure was measured once weekly.Four weeks later,the mRNA expression of PI3K and Akt and Akt protein expression in the liver were measured.Results:After 4 weeks of treatment,the TCM group and Western medicine group had significantly upregulated PI3K mRNA expression in the liver,exhibiting significant differences from that of blank group(P 0.01);the TCM group and Western medicine group also had significantly upregulated Akt mRNA expression in the liver,with significant differences compared with the blank group(P 0.05).The TCM group and Western medicine group had significantly higher Akt protein expression than the blank group(P 0.05).Conclusion:Songling Xuemaikang maintains the normal systolic and diastolic functions of the heart in SHRs possibly by activating the PI3K/Akt signaling pathway,thus inhibiting the adverse effect of hypertension on myocardium.
