摘要
目的:研究四物汤及其活性单体芍药苷和川芎嗪对辐射致血虚证小鼠骨髓细胞核转录因子κB(NF-κB)和转化生长因子β1(TGF-β1)表达的影响,为进一步阐明和比较其补血作用机制提供依据。方法:采用3.5 Gy60Coγ射线全身1次照射,制备血虚证小鼠模型;反向转录-聚合酶链反应检测骨髓细胞NF-κB和TGF-β1mRNA水平。结果:四物汤显著上调血虚证小鼠骨髓细胞NF-κB mRNA,下调TGF-β1mRNA;芍药苷和川芎嗪显著上调血虚证小鼠骨髓细胞NF-κB mRNA,但对TGF-β1mRNA无显著影响。结论:四物汤及活性单体对骨髓细胞NF-κB基因表达的调节和四物汤对TGF-β1基因表达的调节可能是其补血作用的重要机制之一。
Objective:To study the effect of Siwu decoction and its active monomers,paeoniflorin and ligustrazine,on NF-κB and TGF-β1 gene expression in the bone marrow cells of blood deficiency pattern mice induced by radiation and to provide the foundation for further clarifying and comparing the blood enriching mechanism of Siwu decoction and its active monomers.Method:The blood deficiency pattern mice were induced by using a single dose of 3.5 Gy 60Co γ radiation.The mRNA levels of NF-κB and TGF in bone marrow were measured by RT-PCR.Result:Siwu decoction can up-regulate significantly the mRNA level of NF-κB of blood deficiency pattern mice and down-regulate the mRNA level of TGF-β1 in bone marrow cell.Either paeoniflorin or ligustrazine can up-regulate significantly the mRNA level of NF-κB,but has not effect on the mRNA level of TGF-β1.Conclusion:Siwu decoction and its active monomers can regulate NF-κB gene expression in bone marrow cells,but only Siwu decoction can regulate TGF-β1 gene expression in the cells,which probably is one of the important mechanisms of blood proliferation of the decoction.
