摘要
目的探讨丹参注射液对小鼠酒精性肝损伤的保护作用及其防治机制。方法将50只雄性昆明种小鼠分为对照组、模型组(一次性8.0 mL/kg乙醇灌胃造模)和3种不同浓度的丹参处理组(原液组、中浓度组、低浓度组分别给予原液、稀释3倍、稀释5倍丹参注射液腹腔注射10.0 mL/kg,共连续注射8 d),每组各10只。乙醇处理18 h后经摘眼球取血,立即离心分离血清,按血清检查要求保存待检;用全自动生化分析仪检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)与γ-谷氨酰转移酶(GGT),并观察其变化。所有数据均采用方差分析进行处理。结果模型组小鼠ALT、AST与GGT含量与对照组比较,差异有统计学意义(P<0.01);丹参原液组与模型组比较,差异有统计学意义(P<0.01),与对照组比较,差异无统计学意义(P>0.05)。结论高浓度丹参注射液可降低小鼠血清ALT、AST与GGT含量,对小鼠急性酒精性肝损伤有一定的保护作用。
Objective To investigate the protective effects of Danshen Injection on ethanol-induced liver injury in rats and to study its prevention and treatment mechanism.Methods 50 male Kunming mice were randomly divided into to the control group(n=10),model group(n=10,modeling by once ethanol gavage of 8.0 mL/kg body mass) and different concentrations of Danshen treatment groups(original fluid group,middle concentration group and low concentration group,n=10,by intraperitoneal injection of original fluid,3-fold and 5-fold dilution of Danshen Injection 10.0 mL/kg for successive 8 d).After 18 h of ethanol treatment,the eye enucleation was performed for collecting blood sample,the serum was immediately separated by centrifugation and preserved according the serum detection requirements.The fully automatic biochemical analyzer was employed to detect ala nine aminotransferase(ALT),aspartate aminotransferase(AST) and γ-glutamyltransferase(GGT),their changes were observed.All the data were processed by the variance analysis.Results The serum ALT,AST and GGT contents had statstical difference between the model group and the control group(P0.01);the difference between the original fluid group and the model group had statistical significance(P0.01),while the difference had no statistical significance between the original group and control group(P0.05).Conclusion High concentration of Danshen Injection can decrease the serum content of ALT,AST and GGT in rats,and has a certain protective effect on acute ethanol-induced liver injury in rats.
