板蓝根对小鼠实验性肝损伤的保护作用及机制 被引量:2

Protective effects of isatis root on experimental liver injury in mice and its mechanism

摘要 目的探讨板蓝根对四氯化碳(carbon tetrachloride,CCl4)致小鼠肝损伤的保护作用和机制。方法 2013年1—4月选取健康小鼠40只随机分为生理盐水组(A组)、CCl4组(B组)、低剂量板蓝根组(C组)和高剂量板蓝根组(D组)各10只,A、B两组腹腔注射生理盐水10 ml/kg,1次/d;C组腹腔注射板蓝根剂20 ml/kg,1次/d;D组腹腔注射板蓝根剂80 ml/kg,1次/d,四组均注射10 d。末次给药2 h后,B、C、D三组均腹腔注射1 ml/L的CCl4花生油溶液10 ml/kg建立急性CCl4小鼠肝损伤模型。比较四组小鼠肝脏病理组织学变化、肝匀浆中超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)水平、血清ALT、AST水平。计量资料采用t检验,P<0.05为差异有统计学意义。结果空泡变性、肝细胞坏死、炎性细胞浸润情况A组分别为0、0、0只,B组分别为8、10、10只,C组分别为7、6、7只,D组分别为5、4、5只。SOD、MDA水平A组分别为(36.24±3.12)U/mg、(5.20±1.93)nmol/mg,B组分别为(19.21±5.67)U/mg、(8.96±2.65)nmol/mg,C组分别为(25.72±4.35)U/mg、(8.65±2.22)nmol/mg,D组分别为(34.61±4.66)U/mg、(5.98±2.15)nmol/mg。A、B两组比较差异均有统计学意义(均P<0.05);C、D两组与B组比较差异均有统计学意义(均P<0.05)。ALT、AST水平A组分别为(39.06±15.42)、(42.34±16.27)U/L,B组分别为(129.46±39.01)、(144.14±30.51)U/L,C组分别为(100.68±29.36)、(106.54±30.36)U/L,D组分别为(74.25±25.87)、(81.73±20.21)U/L。A、B两组比较差异均有统计学意义(均P<0.05);C、D两组ALT、AST的水平与B组比较差异均有统计学意义(均P<0.05)。结论板蓝根对CCl4诱导的急性肝损伤具有明显的保护作用。 Objective To study the protective effect of Isatis Root on liver injury induced by carbon tetrachloride (CC14) in mice and its mechanism.Methods From January to April in 2013,40 healthy mice were chosen and randomly divided into four groups:NS group(group A),model group(group B),low dose of Isatis Root group(group C) and high dose of Isatis Root group(group D).Normal saline was intraperitoneally administered to both group A and group B in a dose of 10 ml/kg once per day;Isatis root injection was intraperitoneally given to group C in a dose of 20 ml/kg and to group Din a dose of 80 ml/kg once per day.All groups were treated for ten days.Two hours after the last peritoneal administration,CC14 peanut oil solution (1 ml/L) was given in a dose of 10 ml/kg to establish mice models of liver injury. Histopathological changes in the liver, superoxide dismutase (SOD) in liver ho- mogenates,malondialdehyde (MDA) levels,serum levels of ALT and AST were compared in the four groups.Measurement data were processed with t-test,the result of P〈0.05 was considered to be statistically different.Results No cases of vacuolar degener- ation,liver cell necrosis and inflammatory cell infiltration were found in group A; 8 cases of vacuolar degeneration, lO cases of liver cell necrosis and 10 cases of inflammatory cell infiltration were found in group B;7,6 and 7 in group C;5,4 and 5 in group D.The levels o f SOD and MDA were (36.24± 3.12)U/ng and (5.20 ± 1.93)nmol/mg respectively in group A,(19.210 ± 5.67)U/rag and (8.96 ± 2.65)nmol/mg in group B,(25.72 ± 4.35)U/mg and (8.65 ± 2.22)nmol/mg in group C,(34.61 ± 4.66)U/mg and (5.98 ± 2.15)nmol/mgin group D.Significant difference was found between group A and B (P〈0.05).Compared with group B,both group C and D showed an obvious decreasing trend and there was significant difference between them(P〈0.05).The levels of ALT and AST were (39.06 ± 15.42) and (42.34 ± 16.27)U/L in group A,(129.46 ± 39.01) and(144.14 ± 30.51)U/L in group B,(100.68 ± 29.36) and(106.54±30.36)U/L in group C,(74.25 ± 25.87) and (81.73 ± 20.21)U/L in group D.Significant difference was found between group A and B (P〈0.05).The levels of ALT and AST decreased to varying degrees in both group C and D,which were significantly different from those in group B.Conehmion Isatis Root has obvious protective effects on CC14-induced acute liver injury in mice.
