马钱子碱对体外培养的人软骨细胞增殖的影响 被引量:5

摘要 目的观察中药马钱子的提取物马钱子碱(strychnine)对体外培养的人软骨细胞增殖的影响。方法提取、分离人软骨细胞,用甲苯胺蓝染色确定软骨细胞的存在,将第2代软骨细胞以5×104个细胞的密度分别接种于高、中、低剂量组、对照组及硝普钠组,48h后用MTT比色法检测软骨细胞增殖。结果马钱子碱高、低剂量组软骨细胞的增殖率均低于对照组(P<0.05),中剂量组的增殖率高于对照组(P<0.05),并且中、低剂量组能够拮抗一氧化氮对人软骨细胞增殖的抑制作用,其中,中剂量组的作用最强。结论马钱子碱在一定浓度范围内能够有效促进人软骨细胞的增殖。 Objective To investigate the effect of strychnine on proliferation of chondrocyte.Methods The human ehondrocyte was extracted and isolated in vitro. Toluidine blue staining was used to detect the existence of chondrocyte,5x104/mL ehondroeyte of second generation was added to different drug concentrations of strychnine from high dose to low dose respectively and to the control group and to the Sodium nitroprusside group.The proliferation rate of cell was measured by MTT technique after the cell were cultured for 48 hours.Results The proliferation rate of cell growth in high dose group and low dose group were both lower than that in control group(P〈0.05),the inhibition rate of cell growth in medium dose group was higher than that in control group(P〈0.05).In addtion,medium dose group and low dose group can both prevent the inhibtition of chondrocyte growth with nitric oxide,in which medium dose group was more effective.Conclusions Strychnine can promote the proliferation of chondroeyte in proper concentration
