高同型半胱氨酸对小鼠足细胞CD2AP表达的影响

摘要 目的观察α-硫辛酸对高同型半胱氨酸(Hcy)作用下小鼠足细胞胞CD2AP mRNA表达的影响,探讨α-硫辛酸对足细胞的保护作用。方法将体外培养的足细胞分为正常组、Hcy组、Hcy+α-硫辛酸组,培养24、48、72h时分别收集细胞,RT-PCR法检测CD2APmRNA表达。结果 Hcy组,足细胞CD2AP mRNA表达自24h开始下调(P<0.05),且有一定的时效性,72h达到下调高峰(P<0.01);Hcy+α-硫辛酸组,48h时,CD2AP mRNA表达较Hcy组增高(P<0.05),72h仍有上述差异(P<0.05)。结论α-硫辛酸对足细胞有一定的保护作用,可能与其可对抗高同型半胱氨酸下CD2AP mRNA表达下调有关。 Objective To observation the effect of the alpha-lipoic acid on the expression of CD2AP mRNA in mouse podocytes cultured in homocysteine environment,To explore the protective effects of alpha-lipoic acid on podocyte. Methods The podocytes were cultured in high homocysteine and alpha- lipoic acid environment and collected respectively at 24, 48, 72h, the mRNA expression of CD2AP was examined by RT-PCR. Result The expression of CD2AP mRNA was decreased, and there is a certain timelines, alpha lipoic acid can resist the trend. Conclusion Alpha lipoic acid has protective effect on podocytes, and it can resist down-regulating the expression of CD2AP mRNA .
