摘要
主要研究Zn2+对基因工程大肠杆菌发酵生产卤醇脱卤酶的影响。以卤醇脱卤酶活力为响应值,通过单因素试验筛选出显著影响因子Zn2+,并确定其最优浓度为0.0007g·L-1;在50L罐中进行分批发酵试验,比较分析卤醇脱卤酶活力、OD600以及在线参数OUR、CER等的变化趋势,发现Zn2+可显著促进重组大肠杆菌生长,提高生长速率,酶活提高了5.4%。
Studied the influence of Zn2+ on the halohydrin dehalogenase produced by recombinant Escherichia coli P84A/MC1061. First,to regard halohydrin dehalogenation enzyme activity as the response value, determine the significant factor was Zn2+ and the optimal concentration was 0.000 7 g·L -1 by single factor experiments;then,it showed that Zn2+ could significantly promote the growth of recombinant E.Coli and increase the growth rate, and the enzyme activity was a 5.4% increase compared with the control group by comparing and analyzing OD 600 , halohydrin dehalogenase enzyme activity and online parameters OUR,CER and RQ in 50 L batch fermentation experiments.
