摘要
目的研究不同浓度内皮抑素对雌激素影响下的子宫内膜癌系HEC1B细胞增殖的抑制作用。方法以不加药组为对照组,实验1组以不同浓度内皮抑素注射液作用24 h,实验2组以17β-E2 10-8雌激素作用30 min。实验3组以17β-E2 10-8雌激素作用30 min后加入不同浓度内皮抑素注射液作用24 h。应用四甲基偶氮唑盐(MMT)法分别检测各组对细胞增殖的影响。结果实验1组子宫内膜癌系HEC1B随内皮抑素处理浓度增加,吸光度A值明显降低,具有显著性差异(P<0.05)。实验2组用雌激素17β-E2 10-8mol/L作用30 min后,吸光度A值增加。实验3组经过2种药物处理后,内皮抑素浓度增加,吸光度降低明显,与治疗前比较有显著性差异(P<0.05)。结论内皮抑素能明显抑制子宫内膜癌系HEC1B细胞增殖,雌激素能刺激子宫内膜癌系HEC1B细胞增殖,高浓度的内皮抑素能阻抑雌激素对癌细胞的诱导增殖作用。
Objective It is to observe the influence of recombinant human endostatin on proliferation of endometrial cancer cell lines HEC1B induced by estrogens.Methods treating with no drug as control group,various concentrations endostatin(0,12.5,25,50,100,200,400,600 mg / L) as study 1 group and 17β-E2 for 30 minutes as study 2 group and endostatin(0,12.5,25,50,100,200,400,600 mg/L) and 17β-E2 10-8 as study 3 group.The influence of endostatin on proliferation of endometrial cancer cell lines HEC1B was studied by 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazoliumro-mide(MTT) assay.Results In study 1 group after treating with various concentrations of endostatinand,the absorbance of endometrial cancer cell lines HEC1B was significantly decreased(P 0.05).In study 2 group after treating with 17β-E2 10-8 for 30 minutes the absorbance of endometrial cancer cell lines HEC1B was significantly raised(P 0.05).In study 3 group after treating with 17β-E2 10-8 and various concentrations of endostatin,the absorbance of endometrial cancer cell lines HEC1B was significantly decreased(P 0.05).Conclusion Endostatin can inhibit the proliferation of endometrial cancer cell lines HEC1B,17β-E2 10-8 can stimulate the proliferation of endometrial cancer cell lines HEC1B,the proliferation stimulation of estrogen on endometrial cancer cell lines HEC1B can be inhibits by high level endostatin.
