活血化瘀药对COPD模型大鼠气道重塑的干预作用

Intervention of drug for invigorating blood circulation and eliminating stasis on airway remodeling of COPD rat model

目的研究血府逐瘀汤、云南白药对COPD模型大鼠气道重塑的干预作用,揭示活血化瘀药防治COPD模型大鼠气道重塑的机制。方法采用熏烟加气管内滴入内毒素脂多糖的方法复制建立大鼠COPD模型,之后分别给予血府逐瘀汤、云南白药水溶液及生理盐水灌胃,观察大鼠支气管肺组织病理切片的变化,检测大鼠血清中转化生长因子-β1(TGF-β1)、血管内皮生长因子(VEGF)的含量。结果血府逐瘀汤、云南白药对COPD模型大鼠血清中TGF-β1、VEGF均有抑制作用;与云南白药比较,血府逐瘀汤对COPD模型大鼠血清中TGF-β1的抑制作用更大。结论 TGF-β1、VEGF参与COPD气道重塑的病理进程;血府逐瘀汤、云南白药防治COPD气道重塑的机制之一是抑制血清中TGF-β1、VEGF释放;活血化瘀药可以延缓或改善COPD气道重塑的疾病进程,适当配以理气药对提高患者生活质量应该有积极作用。

Objective It is to investigate the influence of Xuefu Zhuyu Decoction and Yunnan Baiyao powder on airway remodeling of chronic obstructive pulmonary disease（COPD） rat model,and reveal the mechanism of promoting blood circulation to remove blood stasis in prevention and cure on airway remodeling of COPD rat model.Methods The rat model of COPD was established by smoking and intratracheal instillation of LPS,then intragastric administration with Xuefu Zhuyu Decoction,Yunnan Baiyao powder aqueous solution and physiological saline respectively.The change of pathological section of lung tissue in rats was observed,and the content of tumor growth factor-β1（TGF-β1） and vascular endothelial growth factor（VEGF） in rat serum was detected.Results The activities of TGF-β1 and VEGF in serum of COPD rat model were markedly inhibited by Xuefu Zhuyu Decoction or Yunnan Baiyao powder,and Xuefu Zhuyu Decoction has more inhibition on TGF-β1 than Yunnan Baiyao power.Conclusion TGF-β1,VEGF is involved in the pathological process of COPD airway remodeling,One of the mechanisms to control airway remodeling of COPD is Xuefu Zhuyu Decoction and Yunnan Baiyao can suppress the release of TGF-β1 and VEGF in the serum.Drug for invigorating blood circulation and eliminating stasis can retard or improve airway remodeling process of COPD disease;appropriate with Qi medicine to improve the life quality of the patients should have a positive effect.
