罗布泊盐湖放线菌分离培养基比较研究 被引量:5

Comparison of Isolation Media for Actinobacteria from LopNur Salt Lake

摘要 目的:认识盐湖环境中放线菌的多样性,挖掘放线菌资源,为今后的开发和利用奠定基础。方法:作者设计出6种培养基,同时补充0~25%NaCl,以高氏一号琼脂培养基(GA)和HV培养基为对照,对采集于新疆罗布泊盐湖的土壤样品进行分离,对获得菌株进行系统发育分析。结果:MM(10个属)、GFA(9个属)、GW1(7个属)、AGG(7个属)和GL3(4个属)培养基得到的放线菌多样性及其新放线菌物种较多,有18个属的放线菌被获得,包括8个放线菌新种。GA和HV培养基仅获得2个属的放线菌,且没有新物种发现。结论:GFA、GW1、AGG、MM和GL3培养基可考虑用于盐湖生态放线菌的分离培养基。另外,不同盐浓度条件下分离培养基获得的放线菌种群结构差异大,建议分离盐湖放线菌时采用多个盐浓度。 Objective:The aim of study was to investigate actinobacterial resources in LopNur Salt Lake and to lay a foundation for furthe- ring to tap. Method: Six media, namely GFA, 307 M, GW1, AGG, MM and GL3 with NaC1 at concentrations ranging between 0% - 25 % (W/V) ,were compared and used to isolate actinobacteria from saline soils in LopNur Lake, China. Phylogenetic diversity of halophilic acti- nomycetes was investigated by using phylogenetic analysis based on 16S rRNA gene sequences. Result:According to test result,these acti- nomycetes isolated from LopNur Salt Lake belong to 18 genera and 8 novel strains ware found. Ten, nine, seven, seven and four genera were isolated using media MM, GFA, GWI, AGG and GL3, respectively. Streptomyces and Nocardiopsis only were isolated by GA medium and HVmedium and no novel species. Conclusion:Five media (GFA, GW1, AGG, MM and GL3 ) were considered as better choice to isolate actinobacteria from salt lake samples in the future. Media with different NaC1 concentrations will impact on isolation result of actinobacterial species.
