PRP/nHA/Co复合材料促进牵张成骨的实验研究

摘要 目的:探讨富含血小板血浆(PRP)、纳米羟基磷灰石(nHA)和胶原(Co)复合材料PRP/nHA/Co促进牵张成骨的效果。方法:取18只健康成年的新西兰大耳兔,建立双侧下颌骨牵张成骨模型。随机分为3组(n=6);A组骨切开处植入PRP/nHA/Co复合膜材料,B组植入nHA/Co复合膜,C组为空白对照组,不放置任何材料。术后4周、8周分别处死动物,取双侧下颌骨,行X线照相,常规切片HE染色,BMP-2及bFGF免疫组织化学染色,光镜下观察。结果:X线、HE组织学观察PRP/nHA/Co组新骨形成均优于其他2组,免疫组织化学观察牵张4周时BMP-2和bFGF表达高于其他2组,8周时表达减弱。结论:PRP/nHA/Co复合材料能够促进牵张成骨。 Objective:To evaluate the effects of PRP/nHA/Co compound in distraction osteogenesis.Methods: Bilateral mandibular distraction osteogenesis model was established in 18 healthy adult New Zealand big ear rabbits.Compound membrane materials of collagen I(Co),nanohydroxyapatie(nHA) and platelet rich plasma(PRP) were prepared as PRP/nHA/Co and nHA/Co,and implanted into the corresponding position of rabbits in group A and group B respectively,in group C no implant was used.After 4 and 8 weeks rabbits were killed respectively,bilateral mandibula were examined by X ray photography,HE staining,BMP-2 and bFGF immunohistochemical staining.Results: New bone formation in PRP/nHA/Co group was superiorer than in other 2 groups.At 4th week of distraction BMP-2 and bFGF expressions in distraction area in PRP/nHA/ Co group were stronger than in other 2 groups,and decreased at the 8th week.Conclusion: PRP/nHA/Co compound material may promote distraction osteogenesis.
