口腔鳞状细胞癌中eIF4E表达的临床意义及预后的相关性分析 被引量:2

摘要 目的:探讨真核细胞翻译起始因子4E(eukaryotic initiation factor 4E,eIF4E)在口腔鳞状细胞癌(OSCC)中的表达及临床意义。方法:采用RT-PCR、Western blot方法及免疫组化SP法检测30例OSCC新鲜组织及112例石蜡包埋组织与30例癌旁正常黏膜组织中eIF4E的表达,分析eIF4E表达与临床病理特征及预后的关系。结果:RT-PCR结果表明eIF4E mRNA在OSCC组织中的表达高于癌旁正常口腔黏膜(P<0.05);Western-blot结果显示eIF4E蛋白在OSCC中高于正常黏膜(P<0.05);多重比较显示,低分化和淋巴结转移病例中eIF4E的表达较高(P<0.05)。Log-Rank生存分析显示eIF4E蛋白低表达组和高表达组的5年生存率分别为63.9%和41.0%(P<0.05);COX多因素分析结果显示,eIF4E蛋白高表达是影响OSCC患者预后的指标之一。结论:eIF4E的高表达与OSCC的发生发展及不良预后密切相关。 Objective:To investigate the expression of eIF4E and its correlation with the clinicopathological parameters and prognosis of oral squamous cell carcinoma(OSCC).Methods: eIF4E mRNA and protein expression in 30 cases of fresh OSCC samples and normal oral mucosa were detected by reverse transcriptase polymerase chain reaction(RT-PCR) and Western-blot respectively.eIF4E protein expression was examined by immunohistochemical(IHC) SP method in 112 cases of paraffin imbedded OSCC samples and 30 of OSCC adjacent tissue.Results: eIF4E mRNA and protein expression in OSCC was higher than in adjacent oral mucosa(P〈0.05),and was related to the pathological differentiation and lymph node metastasis.Log-Rank analysis showed that 5-year survival rate of patients with high and low expression of eIF4E was 41.0% and 63.9% respectively(P〈0.05).The Cox model analysis showed that eIF4E protein expression was related to the poor prognosis of OSCC.Conclusion: eIF4E overexpression is closely related with the occurrence and development of OSCC.
