Effects of GLP-1 Receptor Agonist Exendin-4 on High Glucose Induced Oxidative Stress of Myocytes

摘要 目的:探讨Exendin-4对高糖诱导的心肌细胞氧化应激的影响。方法:通过酶消化法获取乳鼠心肌细胞,在体外高糖暴露条件下,采用不同浓度的Exendin-4处理,MTT法检测细胞的活性,同时运用生化试剂盒测定上清液中丙二醛(MDA)、超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)的水平。结果:与对照组相比,高糖环境下的心肌细胞活性明显减低(P<0.05),MDA含量明显增加(P<0.05),SOD及GSH含量明显减少(P<0.05),而给予Exendin-4预处理后,能逆转上述指标的变化,在浓度为1nmol/L和10nmol/L时具有统计学意义(P<0.05)。结论:高浓度葡萄糖通过氧化应激作用诱导心肌细胞损伤,Exendin-4通过抗氧化作用对心肌细胞起到较好的保护性作用。 Objective: To investigate the effects of glucagon like peptide-1 (GLP-1) receptor agonist on high glucose induced oxidative stress of myocytes. Methods: Neonatal rat ventricular myocytes were prepared by enzymatic dissociation and then cultured with high glucose with or without ex- endin-4, a GLP-1 receptor agonist. Myocardial cell viability was measured by MTT assay kit, and levels of MDA, SOD and GSH were detected using biochemical assay kit. Results: GLP-1 Significantly inhibited the increase in MDA levels induced by high glucose, and the decrease in SOD and GSH levels and cell viability induced by high glucose, especially at the 1 nmol/L and 10 nmol/L concentrations. Conclusion: High concentration of glucose could induce myocardial cell damage and GLP-1 could significantly ameliorate the cardiomyocyte injury induced by high con- centration of glucose by suppressing oxidative stress.
