肝脾调补方对D-氨基半乳糖胺所致小鼠化学性肝损伤的保护作用 被引量:5

Protective Effect of Ganpitiaobu Fang on D-GlaN Induced Liver Injury in Mice

摘要 目的:观察中药肝脾调补方对D-氨基半乳糖胺所致小鼠化学性肝损伤的保护作用并探讨其药理学机制。方法:采用D-氨基半乳糖胺(D-GlaN)建立小鼠急性肝损伤模型,通过肝脾调补方高、中、低剂量灌胃,观察肝脾调补方对各组小鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)及肝组织匀浆超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、一氧化氮(NO)、一氧化氮合酶(NOS)水平的影响。结果:肝脾调补方可明显降低D-氨基半乳糖胺(D-Ga/N)所致肝损伤小鼠血清中ALT、AST水平;降低肝组织匀浆的MDA、NOS、NO的含量,提高肝匀浆SOD、GSH-Px活性。结论:肝脾调补方对D-Ga/N致小鼠肝损伤有保护作用,其作用与抗脂质过氧化、稳定肝细胞膜和线粒体膜有关。 Objective: To study the effects of Ganpitiaobu Fang, a traditional medicine formula, in pro- tecting immunological hepatic injury induced by D-GlaN in mice and study its pharmacological mechanism. Methods: The immunological liver injury rat model was produced by intraperitoneal injection with D-GlaN. Sixty mice were divided into control group, model group, low dose group, middle dose group and high dose group randomly. The levels of plasma alanine amin- otransferase (ALT), aspartate aninotransferase (AST) and liver superoxide dismutase (SOD), malondialdehyde (MDA), glutathione peroxidase (GSH-PX), and nitric oxide (NO) of the five groups were examined. Results: Ganpitiaobu Fang significantly relieved the immunological liver injury in mice by decreasing ALT and AST levels in plasma and MDA content in liver, and eleva- ting the liver contents of SOD and GSH-PX. Conclusion: Ganpitiaobu Fang can obviously protect monce liver from immunological injury, and its effects might be through anti-lipid peroxidation and maintaining the stability of liver cell membrane and mitochondrial membrane.
