摘要
研究定量测定蜂蜜中呋喃唑酮代谢物残留的酶联免疫(ELISA)检测试剂盒的准确性。通过将ELISA样本前处理进行优化,计算添加平均值、回收率及变异系数,并对国标方法和ELISA检测试剂盒测定结果进行t检验。当选择0.1mol/L Tris-HCl为样品稀释液、样本稀释倍数为1、试剂盒灵敏度为0.03μg/kg时,呋喃唑酮代谢物在各蜂蜜中添加0.1~0.3μg/kg的回收率为83%~97%,变异系数均低于5%,试剂盒检测限为0.03μg/kg,ELISA检测试剂盒与国标法测定同一份蜂蜜的t0.05检验结果差异不显著。呋喃唑酮代谢物ELISA检测试剂盒经优化与仪器的符合率为100%,ELISA检测试剂盒测定蜂蜜中呋喃唑酮代谢物存在极高的准确度和精密度。
To evaluate the accuracy of quantitative determination of the metabolite residues of furazoli done in honey by ELISA, the sample pretreatment were optimized and the average, recovery rate and coefficient of variation of differently added concentrations of the same sample were measured. The difference between GB method and ELISA kit was tested. The results showed that, when the sample diluent was 0.1 mol/L Tris-HCl,sample dilution factor was 1, the sensitivity was 0.03 μg/kg, recov ery of furazolidone metabolite of added 0.1-- 0.3 μg/kg in the honey was between 83 % -- 97 %, and the coefficient of variation was less than 5%, kit's limit was 0.03 μg/kg, the results of t005 with the liquid chromatography-tandem mass spectrometry and ELISA were not significant. After optimiza- tion, the coincidence of furazolidone metabolite ELISA kit and liquid chromatography-tandem mass spectrometry was 100%. In conclusion, the determination of furazolidone metabolite in honey with ELISA has high accuracy and precision.
