摘要
为了解甘肃地方桃种质资源的遗传多样性,采用SSR引物对60个甘肃省地方桃品种或类型进行鉴定分析。结果表明:12对引物共扩增出55个条带,其中多态性条带42个,多态性位点百分率为76.53%;平均每对引物产生有效等位基因多态位点数3.25个;位点杂和度(H)为0.537 2~0.757 8,平均为0.647 4;香农指数(I)为0.636 5~1.321 5,平均为0.897 9。UPGMA聚类分析显示,60份材料的相似系数为0.025~0.950,平均为0.630,在相似系数0.675处聚为7个类群。表明甘肃地方桃品种遗传多样性丰富。聚类结果与形态学分类及地理起源基本一致。
Genetic diversity among 60 varieties of Gansu local peach germplasm was analyzed with SSR markers. Fifty five bands were amplified by 12 pairs of primer, of which 42 bands (76.53%) were polymorphic with an average of 3.25 alleles per primer indentified. The heterozygosity (H) ranged from 0. 537 2 to 0. 757 8 with an average of 0. 647 4, and the Shannon index (I) ranged from 0. 636 5 to 1. 321 5 with the average of 0. 897 9. UPGMA cluster analysis showed that the genetic similarity coefficient ranged from 0. 025 to 0. 950, and the clades were grouped to 7 populations at the genetic similarity coefficients of 0. 675. These results showed that genetic diversity difference among tested local peach varieties was relatively high, the cluster result was consistent with the morphological and geographic origin classification.
