Preparation and Immunogenicity of Ploy(Lactide-Co-Glycolide)Microparticles DNA Vaccine against Porcine Reproductive and Respiratory Syndrome

摘要 为增强DNA疫苗的免疫效果,采用溶媒挥发法制备聚乳酸乙醇酸[poly(lactide-co-glycolide)(PLGA)]微粒,将猪繁殖与呼吸综合征(PRRS)DNA疫苗pCI-ORF5吸附到该微粒表面,检测PLGA微粒对DNA的吸附量、体外释放情况以及在小鼠体内的免疫原性。结果表明在6h内PLGA微粒的DNA吸附量可达到0.9%,在体外的释放情况受到CTAB含量、PLGA分子量、PLGA浓度和内水相体积等诸多因素的影响。与裸DNA疫苗同时免疫小鼠后,发现PLGA微粒可显著增强所吸附DNA疫苗诱导的体液免疫和细胞免疫,显示其作为载体递送DNA疫苗方面具有较好的应用前景。 To enhance immune response induced by DNA vaccines, porcine reproductive and respiratory syndrome (PRRS) DNA vaccines pCI-ORF5 was adsorbed on the surface of poly(lactide-co-glycolide) (PLGA) microparticles. The microparticles were prepared using a solvent evaporation method and the adsorption amount, in vitro release profiles and in vivo immunogenicity were evaluated. The results indicated that the DNA adsorbed on PLGA microparticles was about 0.9%in 6 hours, and the release behaviors were influenced by some parameters such as CTAB content, PLGA molecular weight, PLGA concentration, internal aqueous phase volume. The PLGA microparticles with adsorbed DNA induced significantly enhanced humoral and cellular response in comparison to naked DNA in mice. It suggests that PLGA microparticles could be a promising vector for the delivery of DNA vaccines.
