摘要
目的观察不同剂量丹参注射液对小鼠鼠尾表皮颗粒层形成及Th1/Th2细胞因子的影响,探讨丹参治疗银屑病的可能机制。方法用小鼠鼠尾鳞片表皮实验模型,不同组别的小鼠以丹参注射液或甲氨喋呤腹腔注射,观察其对鼠尾表皮颗粒层细胞生成的影响;ELISA法测定各组小鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素-2(IL-2)和4(IL-4)及干扰素-γ(IFN-γ)的水平。结果丹参治疗组可显著促进小鼠鼠尾表皮颗粒层细胞的形成,显著降低小鼠血清TNF-α,IL-2和IFN-γ的水平(P<0.01),其中高剂量组与MTX组差异无统计学意义(P>0.05);丹参治疗组可显著升高小鼠血清IL-4的水平,与MTX组的差异有统计学意义(P<0.01)。结论丹参可能通过调节Thl/Th2细胞因子动态平衡促进小鼠鼠尾表皮颗粒层细胞形成而发挥治疗银屑病的作用。
Objective To explore possible therapeutic mechanism of Salvia miltiorrhiz injection on psoriasis and it's effect on Cytokine Th1/Th2 in mouse psoriasis model. Methods Using mouse tail scale epidermis as exper- imental mode, different groups of mouse were injected Salvia Mihiorrhiz or methotrexate by tra-abdominally and to observe the effects on the formation of epidermal granular layer in mouse tail. The level of TNF-α, IL- 2, IFN-γ/and IL-4 in mouse serum was measured with enzyme linked immunosordent assay (ELISA) meth- od. Results Salvia Miltiorrhiz treated groups could promote greatly the formation of epidermis granular layer in mouse tail and significantly reduced the level of TNF-α, IL-2, IFN-γ in mouse serum ( P 〈 0.01 ). There was no statistically significant difference between MTX group and the higher-dosage of Salvia Miltiorrhiz group ( P 〉 0.05 ). Salvia Mihiorrhiz treated groups could significantly increased the level of IL- 4 in mouse serum(P 〈 0. O1 ). There was significant differences between MTX group and the higher-dosage of Salvia Miltiorrhiz group( P 〈 0.01 ) Conclusion The possible therapeutic mechanism in treating psoriasis with Sal- via Miltiorrhiz might be achieved by modulating the expressions of cytokine Thl/Th2 and promoting greatly the formation of epidermis granular layer in mouse tail.
